TOP STORYResearchers showed here that antagonism of peroxisome proliferator-activated receptor (PPAR)-γ promotes ex vivo expansion of phenotypically and functionally defined subsets of human cord blood HSCs and hematopoietic progenitor cells. [Nat Med] Abstract | |

PUBLICATIONS(Ranked by impact factor of the journal)CRISPR/Cas9 Genome Editing in Human Hematopoietic Stem Cells Scientists provide a detailed protocol for the production, enrichment, and in vitro and in vivo analyses of homologous recombination-targeted HSCs by combining CRISPR/Cas9 technology with the use of rAAV6 and flow cytometry. [Nat Protoc] Abstract TRAF6 Mediates Basal Activation of NF-κB Necessary for Hematopoietic Stem Cell Homeostasis Investigators describe TNF receptor associated factor 6 (TRAF6) as an essential regulator of HSC homeostasis through basal activation of basal nuclear factor κB (NF-κB). Hematopoietic-specific deletion of Traf6 resulted in impaired HSC self-renewal and fitness. [Cell Rep] Full Article | Graphical Abstract The authors found that endothelium-specific transposon mutagenesis in mice promotes hematopoietic pathologies that are both myeloid and lymphoid in nature. Frequently mutated genes included previously recognized cancer drivers and additional candidates, such as Pi4ka, a lipid kinase whose mutation was found to promote myeloid and erythroid dysfunction. [Cell Rep] Full Article | Graphical Abstract While apoptosis and DNA damage response to replicative stress is similar between the control and severe aplastic anemia (SAA)-induced pluripotent stem cell (iPSC)-derived-hematopoietic progenitors, the latter showed impaired proliferation which was not restored by eltrombopag, a drug which has been shown to restore hematopoiesis in SAA patients. [Cell Death Dis] Full Article Applying well-defined surrogate models for the bone marrow niche, live cell imaging techniques, and novel tools in statistical physics, the authors quantified the functionality of two mobilization agents that have been applied in the clinic, NOX-A12 and AMD3100, as compared to a naturally occurring chemokine in the bone marrow, stromal cell-derived factor 1α (SDF1α). [Sci Rep] Full Article Tracking Hematopoietic Precursor Division Ex Vivo in Real Time Researchers confirmed that ecotropic viral integration site 1 (Evi1)-green fluorescent protein (GFP) is a faithful reporter of HSC activity and identified three dividing patterns of HSCs: symmetric renewal, symmetric differentiation, and asymmetric division. They found that the cytokine and growth factor combination promotes symmetric renewal, whereas OP9 stromal cells balance symmetric renewal and differentiation of HSCs ex vivo. [Stem Cell Res Ther] Full Article Investigators showed that ubiquitously expressed minor histocompatibility alloantigens are no relevant target for graft-versus-tumor (GVT) effects. Interestingly, certain ubiquitously expressed MHC alloantigens augmented GVT effects early after transplantation, while others did not. [Bone Marrow Transplant] Abstract CLINICAL RESEARCHScientists analyzed 215 patients with acute myeloid leukemia who underwent allogeneic hematopoietic cell transplant (allo-HCT) in their institution. They limited this retrospective review to patients who received MA conditioning, were in complete remission at the time of transplant and who received a matched sibling transplant, all other patients were excluded to avoid heterogeneity. REVIEWSFrom Hematopoietic Stem Cells to Complex Differentiation Landscapes Evolving views of hematopoiesis have broad implications for an understanding of the functions of adult stem cells, as well as the development of new therapies for malignant and non-malignant hematopoietic diseases. [Nature] Abstract The authors discuss in detail the role that T-cell impairment early after hematopoietic cell transplantation plays in relapse by looking at data from T cell-depleted approaches as well as the clear role that early T-cell recovery has shown in improving outcomes. POLICY NEWSComputer Scientist to Lead French Research Giant; Interim Head Leaves Amid Misconduct Allegations Computer scientist Antoine Petit, 57, is the new head of Europe’s largest research organization. There was an unusual twist to Petit’s appointment, however. A week before the official procedure ended, the French government named Petit interim president of the National Center for Scientific Research, a position that had been held by cell biologist Anne Peyroche since Fuchs’s departure. [ScienceInsider] Editorial Gender Bias Goes Away When Grant Reviewers Focus on the Science But female scientists suffer when their research proposals are judged primarily on the strength of their CVs.
