TOP STORYHGF/R-Spondin1 Rescues Liver Dysfunction through the Induction of Lgr5+ Liver Stem Cells Researchers showed that HGF/Rspo1 induce liver stem cells and rescue liver dysfunction. Carbon tetrachloride treatment promoted both fibrosis and Lgr5+ liver stem cell proliferation, whereas Lgr5 knockdown worsened fibrosis. [Nat Commun] Full Article | |

PUBLICATIONS(Ranked by impact factor of the journal)CBX8 Exhibits Oncogenic Activity via AKT/β-Catenin Activation in Hepatocellular Carcinoma Scientists showed that chromobox 8 (CBX8) expression was increased in hepatocellular carcinoma and correlated with poor outcome in two independent cohorts containing a total of 879 cases. [Cancer Res] Abstract Investigators found SIRT3 protein expression is decreased in tert-butyl hydroperoxide-treated AML12 cells in vitro and primary hepatocytes from CCl4-injured mice in vivo. [Cell Death Dis] Full Article The authors investigated the impact of Caspase-8 in human and murine alcohol-induced apoptosis and in alcoholic liver disease (ALD). They investigated human samples from ALD patients, primary mouse hepatocytes, and hepatocyte-specific Caspase-8 knockout mice in acute and chronic models of ethanol administration. [Cell Death Dis] Full Article miR-9-5p was down-regulated by TGF-β1 in a dose-dependent manner. TGF-β1-induced HSC activation including cell proliferation, α-SMA and collagen expression was blocked down by miR-9-5p [Cell Physiol Biochem] Full Article In Vivo and Ex Vivo Methods of Growing a Liver Bud through Tissue Connection Researchers report a novel transplantation method for liver buds that are grown in vivo involving orthotopic transplantation on the transected parenchyma of the liver, which showed long engraftment and marked growth in comparison to heterotopic transplantation. [Sci Rep] Full Article Scientists investigated the effects of intracellular glyceraldehyde (GA)-derived advanced glycation end-products (GA-AGEs) on human hepatocyte cell death. The accumulation of intracellular GA-AGEs has been associated with the induction of DNA damage and hepatocyte necrotic cell death. [Sci Rep] Full Article Combined Methylome and Transcriptome Analysis during Rat Hepatic Stellate Cell Activation The epigenetic mechanisms accompanying the transition of hepatic stellate cells from a quiescent to an activated state are the focus of this study. The authors investigated the methylome and transcriptome during this process and observed profound changes. While the promoter methylation correlated negatively with gene expression, the gene body methylation revealed no clear correlation. Most genes with altered expression were associated with cell differentiation. [Stem Cells Dev] Abstract Pathways of Production and Delivery of Hepatocyte Exosomes Scientists provide insight regarding the regulation and/or participation of exosome biogenesis or trafficking components in hepatocytes and show that hepatocyte exosomes can mediate therapeutic changes in activated hepatic stellate cells or injured hepatocytes that occur downstream of heparin- or integrin-dependent binding interactions.
