TOP STORYCREG in Hepatocytes Ameliorates Liver Ischemia/Reperfusion Injury in a TAK1-Dependent Manner in Mice In vitro studies were performed using primary hepatocytes isolated from hepatocyte-specific Creg knockout mice that were challenged by hypoxia/reoxygenation insult. These cells exhibited more cell death and inflammatory cytokine production similar to observations in vivo. Further molecular experiments showed that cellular repressor of E1A-stimulated genes (CREG) suppressed MAPK signaling by inhibiting TAK1 phosphorylation. [Hepatology] Abstract

PUBLICATIONS(Ranked by impact factor of the journal)Investigators reported that the RNA-binding protein ELAVL1/HuR played a crucial role in regulating ferroptosis in liver fibrosis. In mice, treatment with sorafenib alleviated murine liver fibrosis by inducing hepatic stellate cell (HSC) ferroptosis. HSC-specific knockdown of ELAVL1 impaired sorafenib-induced HSC ferroptosis in murine liver fibrosis. [Autophagy] Abstract Exposure to poly(ethylene glycol)-poly(lysine) block copolymer-ubenimex conjugate increased the intracellular reactive oxygen species concentration by inhibiting aminopeptidase N enzyme activity, permitting the induction of apoptosis and attenuation of hepatocellular carcinoma cell proliferation. [Oncogene] Abstract | Press Release Researchers reported that a protein named UDP-N-acetylglucosamine pyrophosphorylase-1-like-1 (Uap1l1) was upregulated in the liver of young hepatocyte-specific-Mcl-1 knockout (MKO-hep) mice without any macroscopically detectable tumor nodules and was prominently expressed in the hepatic tumors developed in the aged MKO-hep mice. [Oncogene] Abstract Scientists found that Hsp90β was positively correlated with vasculogenic mimicry and epithelial-mesenchymal transition marker proteins in hepatocellular carcinoma tissues and promoted tube formation, cell migration, and invasion in vitro. [Oncogene] Abstract Investigators found that microRNA-155 was raised in recombinant schistosoma japonicum egg antigen P40 (rSjP40)-treated hepatic stellate cells, and further studies have shown that rSjP40 enhanced microRNA-155 expression by inhibiting STAT5 transcription. Up-regulated microRNA-155 could down-regulate the expression of FOXO3a and then participate in rSjP40-inhibited expression of α-smooth muscle actin and collagen I. [J Cell Mol Med] Full Article Iso-Alpha Acids from Hops (Humulus Lupulus) Inhibit Hepatic Steatosis, Inflammation, and Fibrosis Iso-alpha acids dose-dependently reduced oxidative stress and JNK activation in steatotic primary human hepatocytes, inhibited hepatic stellate cell (HSC) activation, and reduced proliferation and pro-fibrogenic gene expression in already activated HSC in vitro. [Lab Invest] Abstract Unlike c-Myc (Myc) loss, carbohydrate response element-binding protein (ChREBP) loss conferred a proliferative disadvantage to normal murine hepatocytes, as did the combined loss of ChREBP and Myc. The authors observed that Myc and ChREBP cooperatively up-regulated virtually all ribosomal protein genes. [J Biol Chem] Abstract | Full Article Scientists developed a HepaRG-based human liver chimeric mouse model: HepaRG-FRGS. Their experimental results showed that the liver chimerism of the mice was adequate to support chronic hepatitis B virus infection for 24 weeks and to evaluate antivirals. [Emerg Microbes Infect] Full Article Researchers investigated lactoferrin’s (LF) effect on heat shock protein 47 (HSP47) as a potential target for management of liver fibrosis and comparing it with silymarin (SM) in a thioacetamide-induced liver fibrosis model. After six weeks, both LF and SM improved the grade of cirrhosis, reduced collagen fibers deposition, inactivated hepatic stellate cells, significantly decreased elevated liver enzymes, and HSP47. [IUBMB Life] Abstract Ectopic expression of NFATc1 in hepatocellular carcinoma cells inhibited proliferation and colony formation, leading to G1 arrest and induction of apoptosis. The authors demonstrated that NFATc1 increased Fas ligand (FasL) expression by directly binding to its promoter and activated the extrinsic apoptotic pathway. REVIEWSMicroRNAs in Alcoholic Liver Disease: Recent Advances and Future Applications The authors highlight recent findings in miRNA expression and functions as they relate to the pathogenesis of alcoholic liver disease (ALD). Cell-specific distribution of miRNAs, as well as the significance of circulating extracellular miRNAs, is discussed as potential biomarkers. They also evaluate the prospects of miRNA-based therapies in ALD. [J Cell Physiol] Abstract Visit our reviews page to see a complete list of reviews in the hepatic cell research field.
