TOP STORYIn Vitro Expansion of Primary Human Hepatocytes with Efficient Liver Repopulation Capacity Scientists report defined medium conditions that allow 10,000-fold expansion of human hepatocytes (HHs).The proliferating HHs were bi-phenotypic, partially retaining hepatic features while gaining expression of progenitor-associated genes. PUBLICATIONS(Ranked by impact factor of the journal)Inhibition of mTORC2/RICTOR Impairs Melanoma Hepatic Metastasis Researchers focused on the interaction of tumor cells and hepatic stellate cells which play a crucial role in the hepatic microenvironment. In silico analysis revealed increased rapamycin-insensitive companion of mTOR (RICTOR) expression in melanoma cells and tissues and indicated higher expression in advanced melanoma stages and metastases. In vitro, transient RICTOR knock-down via siRNA caused a significant reduction of tumor cell motility. [Neoplasia] Full Article Epigenetic Silencing of TMEM176A Activates Erk Signaling in Human Hepatocellular Carcinoma TMEM176A was highly expressed in SNU387, SNU182, Huh1, and SNU475 cells; reduced expression was observed in HepG2 and PLC/PRF/5 cells; and no expression was found in SNU449, HBXF344, SMMC7721, Huh7, and LM3 cells. [Clin Epigenetics] Full Article A Large-Scale Modeling Approach Reveals Functional Metabolic Shifts during Hepatic Differentiation The authors present an original modelling solution combining metabolic network and omics data to identify modulated metabolic pathways and changes in metabolic functions occurring during differentiation of a human hepatic cell line. They confirm the activation of hepato-specific functionalities and newly evidence modulation of other metabolic pathways, which could not be evidenced from transcriptomic data alone. [J Proteome Res] Abstract Hepatic stellate cell (HSC)-conditioned medium (CM) significantly enhanced the proliferation, motility, invasion, prominent epithelial-mesenchymal transition activation and decreased apoptosis of heat-exposed residual hepatocellular carcinoma cells. These increased malignant phenotypes were markedly attenuated by neutralizing periostin in HSC-CM. [J Transl Med] Full Article LGR4 Protects Hepatocytes from Injury in Mouse Scientists demonstrated that in adult liver, LGR4 was expressed in hepatocytes and responds to Rspo1 with internalization. Rspo1 was responsive to various nutritional states and to mTOR signaling. Activation of LGR4 by Rspo1 significantly reduced TNFα-induced cell death, and levels of NFκB-p65 and caspase 3 in cultured hepatocytes. [Am J Physiol Gastrointest Liver Physiol] Abstract Researchers found that reactive oxygen species can promote lipid droplet formation in hepatocytes by promoting perilipin2 expression. They used different concentrations of hydrogen peroxide to treat HepG2 cells and found that the number of lipid droplets in the cells increased, however also that this effect was dose-independent. [Int J Mol Sci] Full Article Plac9 is a novel protein that is localized to the cytoplasm and may be secreted through the classic endoplasmic reticulum-Golgi route. The overexpression of Plac9 inhibits cell growth and induces G2/M phase arrest. [Biosci Rep] Abstract Transport of Salvianolic Acid B via the Human Organic Anion Transporter 1B1 in the Liver Investigators explored the uptake features of salvianolic acid B (SAB) in HepG2 cells and the effect of rifampicin on uptake. They explored the effects of SAB on the uptake of pitavastatin in HepG2 cells. Finally, they established an HEK239T-OATP1B1 cell model to confirm whether OATP1B1 mediated the transport of SAB.
