    Impaired NK-Mediated Regulation of T-Cell Activity in Multiple Sclerosis Is Reconstituted by IL-2 Receptor Modulation
    Investigating the peripheral blood and cerebrospinal fluid of multiple sclerosis (MS) patients treated with natalizumab revealed that transmigration of this subset depends on the α4β1 integrin very late antigen-4. Although no MS-related changes in the migratory capacity of natural killer (NK) cells were observed, NK cells derived from patients with MS exhibit a reduced cytolytic activity in response to antigen-activated CD4+ T cells.     Human Macrophages Induce CD4+Foxp3+ Regulatory T Cells via Binding and Re-Release of TGF-β
    As a prerequisite for eventual translation of immunosuppressive macrophage (M2) therapy into clinical settings the authors studied the induction, stability and mechanism of generation of human induced Tregs by M2 macrophages generated with interleukin-4/interleukin-10/transforming growth factor-β (TGF-β).     BCR–ABL-Specific CD4+ T-Helper Cells Promote the Priming of Antigen-Specific Cytotoxic T Cells via Dendritic Cells

    Redirecting T-Cell Specificity to EGFR Using mRNA to Self-Limit Expression of Chimeric Antigen Receptor
    Researchers evaluated expression of an epidermal growth factor receptor (EGFR)-specific chimeric antigen receptor coded from introduced mRNA in human T cells numerically expanded ex vivo to clinically significant numbers through coculture with activating and propagating cells derived from K562 preloaded with anti-CD3 antibody.     Increased Th17 Cells and IL-17A Exist in Patients with B Cell Acute Lymphoblastic Leukemia and Promote Proliferation and Resistance to Daunorubicin through Activation of Akt Signaling
    Scientists assessed and observed a significantly increased frequency of Th17 cells and a drastically decreased frequency of Th1 cells in peripheral blood mononuclear cells and bone marrow mononuclear cells from newly diagnosed B-cell acute lymphoblastic leukemia patients compared with healthy donors.     Promoter H3K4 Methylation Dynamically Reinforces Activation-Induced Pathways in Human CD4 T Cells
    Researchers have detailed the dynamics of genome-wide promoter H3K4me2 and H3K4me3 over a time course during activation of human naive and memory CD4 T cells. They demonstrated that changes to H3K4 methylation occur relatively late after activation and reinforce activation-induced upregulation of gene expression, affecting multiple pathways important to T-cell activation, differentiation and function.     Aberrant Expression of B7-H4 Correlates with Poor Prognosis and Suppresses Tumor-Infiltration of CD8+ T Lymphocytes in Human Cholangiocarcinoma
    The authors investigated the expression of B7-H4 in patients with cholangiocarcinoma and its association with patient prognosis. The correlation between B7-H4 expression and CD4+ and CD8+ tumor-infiltrating lymphocytes was also investigated. [Oncol Rep] Abstract
