TOP STORYResearch Identifies Novel Unconventional Type of Immune Cell Capable of Fighting Viral Infections Researchers identified a novel unconventional type of immune cell capable of fighting viral infections. Specifically the research defined a subset of ‘unconventional V-delta-2 lymphocytes’, which are a type of gamma delta T cell – an ancient class of immune cell that has been relatively poorly understood. PUBLICATIONS(Ranked by impact factor of the journal)The authors found that glycerol monolaurate-treated human T cells were less adherent than untreated T cells and did not form actin ring structures but instead developed numerous inappropriate actin-mediated filopodia. [Sci Signal] Abstract Investigators stimulated human blood monocytes with agonists for TLR4 (LPS), TLR3 and TLR7/8 and subsequently challenged them to low doses of endotoxin. The different TLR agonists promoted distinct inflammatory signatures in monocytes. [Cell Death Dis] Full Article IL-3 but not Monomeric IgE Regulates FcεRI Levels and Cell Survival in Primary Human Basophils As basophils and mast cells share many biological commonalities the authors assessed the role of monomeric IgE binding and interleukin-3 (IL-3) signaling in FcεRI regulation and cell survival of primary human basophils. [Cell Death Dis] Full Article In vitro treatment with infliximab greatly promoted Crohn’s disease (CD) CD4+ T cells to express IL-22, which was inhibited by recombinant human TNF-α converting enzyme, indicating that reverse signaling through binding to membrane-bound tumor necrosis factor (TNF) mediates anti-TNF-induced IL-22 expression of CD CD4+ T cells. [Inflamm Bowel Dis] Abstract Specific Features of Human Monocytes Activation by Monophosphoryl Lipid A Researchers deciphered the mechanisms of production of pro- and anti-inflammatory cytokines by adherent human blood mononuclear cells activated by lipopolysaccharide or monophosphoryl lipid A. [Sci Rep] Full Article Scientists analyzed PBMCs from 29 individuals representing the haplotypes containing each of the significantly associated signals. STAT4 phosphorylation was evaluated by phospho-flow cytometry after CD3/CD28 activation of cells followed by IL-12 stimulation. [Sci Rep] Full Article Anti-tumor efficacy of human epidermal growth factor receptor 2 (HER2)-BBz-chimeric antigen receptor (CAR) T cells in vitro was performed using cytokine release and immune cytotoxicity assays compared to control CD19 CAR T cells. [J Immunother Cancer] Full Article Investigators evaluated the ability of exogenous serotonin to modulate the T-cell behavior of patients with multiple sclerosis, a demyelinating autoimmune disease mediated by Th1 and Th17 cytokines. [Eur J Immunol] Abstract Peripheral blood mononuclear cells were isolated from blood of SLE patients as well as healthy donors. The proportion of Th cell subsets classified from cell surface markers is detected by flow cytometry. INDUSTRY NEWSAtara Biotherapeutics, Inc. announced the company has expanded its collaboration with Memorial Sloan Kettering Cancer Center to develop the next generation of genetically engineered chimeric antigen receptor (CAR) T immunotherapies. [Atara Biotherapeutics, Inc.] Press Release Cancer Center at BIDMC Launches Immunotherapy Institute The Cancer Center at Beth Israel Deaconess Medical Center (BIDMC) announced the launch of a new clinical and research institute that will build on BIDMC’s long history of leadership in immunotherapy and cell therapeutics. The Immunotherapy Institute will harness the potential of immunotherapy as it revolutionizes the way cancer and ultimately other diseases are treated and diagnosed. [Beth Israel Deaconess Medical Center] Press Release Novartis announced the FDA has approved Kymriah® suspension for intravenous infusion for its second indication – the treatment of adult patients with relapsed or refractory (r/r) large B-cell lymphoma after two or more lines of systemic therapy including diffuse large B-cell lymphoma (DLBCL), high grade B-cell lymphoma and DLBCL arising from follicular lymphoma. POLICY NEWSCancer-Killing Viruses Show Promise — and Draw Billion-Dollar Investment Pharmaceutical giant Johnson & Johnson announced that it would pay up to US$1 billion to acquire a company that makes cancer-killing viruses. It was a striking show of support for a still-unproven treatment, but the bid is just the latest sign that industry and academics are warming to the approach. [Nature News] Editorial Postdoc Hopes Pennsylvania Voters Will Help Her Re-Engineer How to Run for Congress A postdoctoral researcher at the University of Pennsylvania who designs proteins for possible use in treating cancer, Sheehan has ignored conventional wisdom about the importance of raising tons of money to spread her message that she’s “a mother, a bioengineer, [and a] proud progressive.” Instead, she’s urging scientists and others in the community to offer their considerable skills to work for candidates they support. [ScienceInsider] Editorial Basic Scientists Still Feel Pinch of New NIH Clinical Trial Policy Basic researchers who study the brain and human behavior thought lawmakers had come to their rescue in March by blocking the National Institutes of Health (NIH) from redefining their studies as clinical trials. But NIH officials are still pushing ahead with new requirements that scientists say make no sense and will cripple their research. [ScienceInsider] Editorial Petition Asks National Academy of Sciences to Boot Sexual Harassers An online petition calling on the National Academy of Sciences to revoke memberships bestowed on people sanctioned for sexual harassment has garnered more than 250 signatures since it was posted on May 1.
