A Human Apolipoprotein L with Detergent-Like Activity Kills Intracellular Pathogens

Scientists screened 19,050 human genes by CRISPR-Cas9 mutagenesis and identified interferon-gamma–induced apolipoprotein L3 as a potent bactericidal agent protecting multiple non–immune barrier cell types against infection.

Non-Terminally Exhausted Tumor-Resident Memory HBV-Specific T Cell Responses Correlate with Relapse-Free Survival in Hepatocellular Carcinoma

Using highly multiplexed peptide-MHC tetramer staining of unexpanded cells from blood, liver, and tumor tissues from 46 hepatocellular carcinoma patients, researchers detected 91 different antigen-specific CD8+ T cell populations targeting hepatitis B virus (HBV), neoantigen, tumor-associated, and disease-unrelated antigens.

BATF and IRF4 Cooperate to Counter Exhaustion in Tumor-Infiltrating CAR T Cells

Overexpression of basic leucine zipper ATF-like transcription factor (BATF) in CD8+ T cells expressing a chimeric antigen receptor (CAR) promoted the survival and expansion of tumor-infiltrating CAR T cells, increased the production of effector cytokines, and decreased the expression of inhibitory receptors.

Adenovirus Vector Vaccination Reprograms Pulmonary Fibroblastic Niches to Support Protective Inflating Memory CD8+ T Cells

The authors showed that clinically applied adenovirus vectors preferentially targeted fibroblastic stromal cells in cultured human tissues.

Alternative Pathways for the Development of Lymphoid Structures in Humans

In humans, JAK3 and γc deficiencies resulted in severe combined immunodeficiency characterized by an absence of T cells, natural killer cells, innate lymphoid cells, and presumably lymphoid tissue inducer cells.

    Abstract

Polygenic Risk Score and Risk of Monoclonal B-Cell Lymphocytosis in Caucasians and Risk of Chronic Lymphocytic Leukemia (CLL) in African Americans

A polygenic-risk-score (PRS) of 41 CLL-susceptibility variants has been found to be associated with CLL risk among individuals of European-ancestry. Scientists evaluated these variants, the PRS, and environmental factors for monoclonal B-cell lymphocytosis risk.

TNFα Increases Tyrosine Hydroxylase Expression in Human Monocytes

The authors reported the development of a highly sensitive and reproducible Bio-ELISA to quantify picogram levels of tyrosine hydroxylase in multiple model systems. They applied this assay to monocytes isolated from blood of persons with Parkinson’s disease and to age-matched, healthy controls.

Single-Cell Transcriptome and TCR Profiling Reveal Activated and Expanded T Cell Populations in Parkinson’s Disease

Scientists performed single-cell transcriptome and TCR sequencing, and conducted integrative analyses to decode composition, function and lineage relationship of T cells in the blood and cerebrospinal fluid of Parkinson’s disease.

S100A9 in Adult Asthmatic Patients: A Biomarker for Neutrophilic Asthma

The authors investigated the mechanism by which S100 calcium-binding protein A9 (S100A9) was involved in neutrophil activation, neutrophil extracellular trap-induced airway inflammation, and macrophage polarization in neutrophilic asthma.

Adversity in Early Life and Pregnancy Are Immunologically Distinct from Total Life Adversity: Macrophage-Associated Phenotypes in Women Exposed to Interpersonal Violence

Researchers examined whether retrospective reports of adversity during childhood or pregnancy were associated with inflammatory imbalance, in an ethnically diverse cohort of 53 low-income women seeking family-based trauma treatment following exposure to interpersonal violence.

CD40 Stimulation as a Molecular Adjuvant for Cancer Vaccines and Other Immunotherapies

The rationale and early immunobiology of CD40 as a master regulator of dendritic cell activation is reviewed, with further contextualization and appreciation for the role of CD40 stimulation not only in cancer vaccines but also in other contemporary immune-oncology approaches.
