T Cell Receptor Recognition of Hybrid Insulin Peptides Bound to HLA-DQ8

HLA-DQ8-HIP tetramer+ T cells from the peripheral blood of a type I diabetes (T1D) patient were characterized by repeated TRBV5 usage, which matched the TCR bias of CD4+ T cells reactive to the hybrid insulin peptide isolated from the pancreatic islets of a patient with T1D.

Functional Th1-Oriented T Follicular Helper Cells That Infiltrate Human Breast Cancer Promote Effective Adaptive Immunity

Scientists found that CD4+ Tfh tumor infiltrating lymphocyte (TIL), CD8+ TIL and TIL-B, co-localizing in tertiary lymphoid structures, all expressed the CXCL13 receptor CXCR5.

CD4+ T Cells in the Lungs of Acute Sarcoidosis Patients Recognize an Aspergillus nidulans Epitope

The authors screened related Löfgren’s syndrome-specific TCRs for antigen specificity and identified a peptide derived from NAD-dependent histone deacetylase hst4 of Aspergillus nidulans that stimulated the CD4+ T cells in an HLA-DR3–restricted manner.

Up-Regulation of C/EBPα Inhibits Suppressive Activity of Myeloid Cells and Potentiates Antitumor Response in Mice and Cancer Patients

Investigators conducted a Phase I trial in 36 patients with hepatocellular carcinoma who were given therapeutic C/EBPα saRNA (MTL-CEBPA) as either neoadjuvant or adjuvant treatment. MTL-CEBPA treatment in those patients caused a marked decrease in peripheral blood monocytic myeloid-derived suppressor cell numbers.

    Abstract

Targeting the Atf7ip-Setdb1 Complex Augments Antitumor Immunity by Boosting Tumor Immunogenicity

To identify epigenetic regulators of tumor antigen expression, scientists established a transplantable syngeneic tumor model of immune escape with silenced antigen expression and used this system as a platform for a CRISPR-Cas9 suppressor screen for genes encoding epigenetic modifiers.

Cooperativity Mediated by Rationally Selected Combinations of Human Monoclonal Antibodies Targeting the Henipavirus Receptor Binding Protein

Researchers isolated a panel of human monoclonal antibodies from the B cells of an individual with prior exposure to equine Hendra virus vaccine, targeting distinct antigenic sites.

High-Grade Serous Ovarian Tumor Cells Modulate NK Cell Function to Create an Immune-Tolerant Microenvironment

Investigators applied mass cytometry and uncover decidual-like natural killer cell subpopulations in newly diagnosed high-grade serous carcinoma samples that correlated with both tumor and transitioning epithelial-mesenchymal cell abundance.

Targeted T Cell Receptor Gene Editing Provides Predictable T Cell Product Function for Immunotherapy

By studying 51 different T cell receptors (TCRs), researchers showed that conventional genetic engineering by viral transduction led to variable TCR expression and functionality as a result of variable transgene copy numbers and untargeted transgene integration.
