    Functional HPV-Specific PD-1+ Stem-Like CD8 T Cells in Head and Neck Cancer

Researchers studied CD8 T cells in patients with human papillomavirus (HPV)-positive head and neck cancer and identified several epitopes derived from HPV E2, E5 and E6 proteins that allowed them to analyze virus-specific CD8 T cells using major histocompatibility complex class I tetramers.

The Immunostimulatory RNA RN7SL1 Enables CAR-T Cells to Enhance Autonomous and Endogenous Immune Function

Investigators engineered CAR-T cells to deliver RN7SL1, an endogenous RNA that activates RIG-I/MDA5 signaling. RN7SL1 promoted expansion and effector-memory differentiation of CAR-T cells.

Effector Differentiation Downstream of Lineage Commitment in ILC1s Is Driven by Hobit across Tissues

Through multiplexed single-cell mRNA sequencing, the authors identified cKit+CD127hiTCF-1hi early differentiation stages of T-bet+ innate lymphoid cells.

    Abstract

A Reservoir of Stem-Like CD8+ T Cells in the Tumor-Draining Lymph Node Preserves the Ongoing Anti-Tumor Immune Response

The authors found that a small number of TCF1+ tumor-specific CD8+ T cells were present in lung tumors throughout their development.

Integrin-αV-Mediated Activation of TGF-β Regulates Anti-tumour CD8 T Cell Immunity and Response to PD-1 Blockade

Using multiplex immunofluorescence staining in cohorts of anti-PD-1 and anti-PD-L1-treated lung cancer patients, scientists showed that decreased expression of cancer cell αV was associated with improved immunotherapy-related, progression-free survival.

PHF6 and JAK3 Mutations Cooperate to Drive T-Cell Acute Lymphoblastic Leukemia Progression

The average survival time was shorter in patients with JAK/STAT and PHF6 comutation than that in other patients, suggesting a potential role of PHF6 in leukemia progression.

Chronic Stress Primes Innate Immune Responses in Mice and Humans

Monocytes from stressed mice and humans exhibit a characteristic inflammatory transcriptomic signature and are hyperresponsive upon stimulation with Toll-like receptor ligands.

Immunomodulatory Effect of NEDD8-Activating Enzyme Inhibition in Multiple Myeloma: Upregulation of NKG2D Ligands and Sensitization to Natural Killer Cell Recognition

Investigators found that pharmacologic inhibition of neddylation using a small-molecule inhibitor, MLN4924/Pevonedistat, increased the expression of the natural killer (NK) cell-activating receptor NKG2D ligands MICA and MICB on the plasma membrane of different multiple myeloma cell lines and patient-derived plasma cells, leading to enhanced NK cell degranulation.

Mitogen-Activated Protein Kinase Signaling in Childhood Asthma Development and Environment-Mediated Protection

Scientists aimed to investigate the role of MAPK signaling in childhood asthma and its environment-mediated protection, including a representative selection of 232 out of 1062 children from two cross-sectional cohorts and one birth cohort study.

Regulation of Antitumor Immunity by Inflammation-Induced Epigenetic Alterations

The authors discuss the role of inflammation in initiating epigenetic alterations in immune cells, cancer-associated fibroblasts, and cancer cells and suggest how and when epigenetic interventions can be combined with immunotherapies to improve therapeutic outcomes.
