TOP STORYScientists Discover How Gene Mutation Triggers Immune Disease Scientists discovered how a gene mutation affects T cell function to promote immune disorders and then tested a treatment based on the discovery—successfully fixing donated immune cells from a 16-year-old boy with an abnormally low level of white blood cells called lymphopenia. PUBLICATIONS(Ranked by impact factor of the journal)IL-6 Receptor Blockade Corrects Defects of XIAP-Deficient Regulatory T Cells The authors showed that mouse Xiap−/− regulatory T (Treg) cells and human XIAP-deficient Treg cells are defective in suppressive function. The Xiap−/− Treg cell defect was linked partly to decreased SOCS1 expression. [Nat Commun] Full Article CEACAM1 Controls IL-2-Dependent Regulatory T Cell Induction in Immune-Mediated Hepatitis Carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1)-/-CD4+ T cells showed impaired IL-2-mediated signal transducer and activator of transcription 5 phosphorylation, which correlated with a failure of naïve CEACAM1-/-CD4+ T cell to convert into regulatory T cells in vitro. [Hepatology] Abstract Cell depletion studies revealed that NK cells and CD8+ T cells were required for intracranial anti–PD-1/anti–CTLA-4 efficacy. Rather than enhancing CD8+ T cell activation and expansion within intracranial tumors, PD-1/CTLA-4 blockade dramatically increased the trafficking of CD8+ T cells to the brain. [Proc Natl Acad Sci USA] Abstract The authors scrutinized platelet responses to systemic immune complexes (ICs) in the absence of tissue and endothelial wall injury. Platelet activation by circulating ICs through a mechanism requiring expression of platelet Fcγ receptor IIA resulted in the induction of systemic shock. [Proc Natl Acad Sci USA] Abstract | Press Release Treatment with TNCscTNF80 increased arthritis-induced myelopoiesis. Little effect was seen on the infiltration rate of inflammatory immature myeloid cells and on the reduction of lymphoid cells in secondary lymphoid organs. Upon treatment, frequency of regulatory T cells in the CD4+ T-cell population was increased in both spleen and inguinal lymph nodes. [Cell Mol Immunol] Abstract MLL1 Promotes IL-7 Responsiveness and Survival during B Cell Differentiation Using a Rag1-cre mouse knock-in to selectively delete Mll1 in developing lymphocytes, scientists showed that B cell, but not T cell, homeostasis depends on MLL1. Mll1−/− B progenitors fail to differentiate efficiently through the pro- to pre-B cell transition, resulting in a persistent reduction in B cell populations. [J Immunol] Abstract The authors explored the role of guanylate binding protein 1, an intracellular anti-microbial protein, in regulating polarization, metabolic reprogramming, and cellular aging of macrophages. [Sci Rep] Full Article Researchers investigated the roles of B cell scaffold protein with ankyrin repeats 1 (BANK1) in collagen-induced arthritis and the interaction between B cells and induced T regulatory cells. [Arthritis Res Ther] Full Article Investigators tested the effects of collagen-peptide ingestion on allergic response and the effects of collagen-derived oligopeptides on CD4+ T-cell differentiation. REVIEWSEpigenetic Control of CD8+ T Cell Differentiation Researchers review the epigenetic processes that direct CD8+ T cell differentiation and function. They focus on epigenetic modification of DNA and associated histones at genes and their regulatory elements. [Nat Rev Immunol] Abstract Epigenetic Regulation in B-Cell Maturation and Its Dysregulation in Autoimmunity The authors summarize current research progress in epigenetic modifications contributing to B-cell activation and differentiation, especially under autoimmune conditions such as lupus, rheumatoid arthritis and type 1 diabetes. POLICY NEWSBulgaria in the Cold as European Union Freezes Its Funding European Union science ministers are due to meet in their bloc’s poorest member state — Bulgaria — to discuss future EU research policy. For the host nation, it was supposed to be a chance to showcase its ambitious plans to boost economic growth by attracting international research institutes to the country. [Nature News] Editorial Kid Co-Authors in South Korea Spur Government Probe The South Korean government is expanding an investigation into researchers who named their children as co-authors on papers. The extended probe comes after a government report identified 82 academic papers on which authors had named their children or relatives — many of them in middle or high school — as co-authors on the publications.
