Immune System Key: New Study Questions Earlier Conclusions about the Kinetics of T Cell Receptors
Based on two techniques that mechanically study receptors as they operate on T cell membranes, the findings could lead to a reevaluation of prior conclusions. [Press release from Georgia Institute of Technology discussing online prepublication in Nature]

    SCIENCE NEWS

Researchers Use Nanoparticle “Vaccine” to Cure Type 1 Diabetes in Mice
Using an innovative nanotechnology-based “vaccine,” researchers were able to successfully restore normal blood sugar in mice with type 1 diabetes, and also slow the onset of diabetes in mice at risk for the disease. [Press release from Juvenile Diabetes Research Foundation discussing online prepublication in Immunity]

University of Central Florida Professor Harnesses Plant ‘Power’ to Fight Hemophilia
Treating hemophilia can sometimes be deadly because many patients suffer fatal allergic reactions to the expensive protein that doctors use to make their blood clot. Now researchers have devised a way that could help patients develop a tolerance to the therapeutic protein before they seek treatment to prevent the potentially deadly reactions. [Press release from University of Central Florida discussing online prepublication in Proceedings of the National Academy of Sciences U S A]

    CURRENT PUBLICATIONS

Conversion of Adult Pancreatic Alpha-Cells to Beta-Cells After Extreme Beta-Cell Loss
Researchers show beta-cell regeneration in a transgenic model of diphtheria-toxin-induced acute selective near-total beta-cell ablation. [Nature]

The Kinetics of Two-Dimensional TCR and pMHC Interactions Determine T Cell Responsiveness
2D data suggest rapid antigen sampling by T cells and serial engagement of a few agonist peptide-major histocompatibility complexes (pMHCs) by T cell receptor (TCR)s in a large self pMHC background. Thus, the cellular environment amplifies the intrinsic TCR-pMHC binding to generate broad affinities and rapid kinetics that determine T cell responsiveness. [Nature]

Protein Kinase C-Theta Mediates Negative Feedback on Regulatory T Cell Function
Inhibition of PKC-theta protected regulatory T cell (Treg) from inactivation by TNF-alpha, restored activity of defective Treg from rheumatoid arthritis patients, and enhanced protection of mice from inflammatory colitis. [Science]

Induction of Lymphoidlike Stroma and Immune Escape by Tumors That Express the Chemokine CCL21
Researchers suggest that by altering the tumor microenvironment, CCL21-secreting tumors shift the host immune response from immunogenic to tolerogenic, thereby facilitating tumor progression. [Science]

T Helper Type 1 and 17 Cells Determine Efficacy of Interferon-Beta in Multiple Sclerosis and Experimental Encephalomyelitis
Investigators have found congruence in outcome between responses to interferon-beta in experimental autoimmune encephalomyelitis and relapsing-remitting multiple sclerosis. [Nat Med]

Reversal of Autoimmunity by Boosting Memory-like Autoregulatory T Cells
Researchers show that progression to autoimmune diabetes results in the conversion of naive low-avidity autoreactive CD8+ T cells into memory-like autoregulatory cells that can be expanded in vivo with nanoparticles coated with disease-relevant peptide-major histocompatibility complexes. [Immunity]

    TRAF6 Inhibits Th17 Differentiation and TGF-Beta-Mediated Suppression of IL-2
Here researchers report that CD4+ T cells lacking the molecular adaptor TNF receptor-associated factor 6 (TRAF6) exhibit a specific increase in Th17 differentiation in vivo and in vitro. [Blood]

Toll-Like Receptor Induced Changes in Glycolytic Metabolism Regulate Dendritic Cell Activation
These data pinpoint toll-like receptor-mediated metabolic conversion as essential for dendritic cell maturation and function, and reveal it as a potential target for intervention in the control of excessive inflammation and inappropriately regulated immune responses. [Blood]

Plasmacytoid Dendritic Cells Resident in Human Thymus Drive Natural Regulatory T Cell Development
Here researchers report that plasmacytoid dendritic cells (pDCs) resident in the human thymus, when activated with CD40L plus IL-3, efficiently promoted the generation of CD4+CD25+Foxp3+ natural regulatory T cells from autologous thymocytes. [Blood]

Lentiviral-Mediated Foxp3 RNAi Suppresses Tumor Growth of Regulatory T Cell-Like Leukemia in a Murine Tumor Model
Inhibiting Foxp3 gene expression by shRNAs effectively decreases tumor growth of regulatory T cell-like leukemia. [Gene Ther]
