SCIENCE NEWS When Helper Cells Aren’t Helpful Current research suggests that T helper-type 1 (Th1) cells, previously thought to mediate autoimmunity, may actually inhibit the development of experimental immune encephalomyelitis, a mouse model of multiple sclerosis, by suppressing Th17 cells. [Press release from Biocompare discussing online prepublication in the American Journal of Pathology] Transplant Tolerance ‘Signature’ Identified A team of researchers have identified a ‘full set’ of immunological markers in the blood, which could be used to predict whether an individual’s kidney transplant will be a long term success or whether it will fail. CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) The T Helper Type 2 Response to Cysteine Proteases Requires Dendritic Cell-Basophil Cooperation via ROS-Mediated Signaling Here researchers demonstrate that cysteine protease-induced TH2 responses occur via ‘cooperation’ between migratory dermal dendritic cells and basophils positive for interleukin 4. [Nat Immunol] A Molecular Classifier for Predicting Future Graft Loss in Late Kidney Transplant Biopsies To assess the kidney upon developing signs of renal dysfunction or proteinuria, a “for-cause” biopsy is performed, but this provides little indication as to which recipients will go on to organ failure. In an attempt to identify molecules that could provide this information, researchers used micorarrays to analyze gene expression in 105 for-cause biopsies taken between 1 and 31 years after transplantation. [J Clin Invest] Grb2 Functions at the Top of the T Cell Antigen Receptor-Induced Tyrosine Kinase Cascade to Control Thymic Selection Here researchers show that conditional ablation of Grb2 in thymocytes severely impairs both thymic positive and negative selections. [Proc Natl Acad Sci U S A] B Lymphoid Cells with Attributes of Dendritic Cells Regulate T Cells Via Indoleamine 2,3-Dioxygenase Here researchers show that indoleamine 2,3-dioxygenase-competent cells express the B lineage commitment factor Pax5 and surface immunoglobulins. [Proc Natl Acad Sci U S A] Immature Immunosuppressive CD14+HLA-DR(-/low) Cells in Melanoma Patients are Stat3(hi) and Overexpress CD80, CD83, and DC-Sign In this study, researchers have examined circulating CD14+HLA-DR(-/low) myeloid-derived suppressor cells in 34 advanced malignant melanoma patients. [Cancer Res] Immune Infiltration of Spontaneous Mouse Astrocytomas Is Dominated by Immunosuppressive Cells from Early Stages of Tumor Development Overall, the results show an early defective endogenous immune response to gliomas, and local accumulation of immunosuppressive cells at the tumor site. [Cancer Res] Selective Depletion of CD4+CD25+Foxp3+ Regulatory T Cells by Low-Dose Cyclophosphamide Is Explained by Reduced Intracellular ATP Levels In this report, researchers provide evidence that ATP, the energy molecule and signal element, accounts for the selective depletion of regulatory T cells by low-dose cyclophosphamide. [Cancer Res] Control of T Cell Reactivation by Regulatory Qa-1-Restricted CD8+ T Cells These experiments provide new insight into a CD8+ T cell compartment that regulates the immediate reactivation of conventional T cells and natural killer T cells.
