CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) Functionally Defective Germline Variants of Sialic Acid Acetylesterase in Autoimmunity Functionally defective SIAE rare and polymorphic variants represent a strong genetic link to susceptibility in relatively common human autoimmune disorders. [Nature] Viral Infection Triggers Central Nervous System Autoimmunity via Activation of CD8+ T Cells Expressing Dual TCRs Here researchers found that CD8+ myelin basic protein (MBP)-specific T cell tolerance was broken and autoimmunity was induced by infection with a virus that did not express MBP cross-reactive epitopes and did not depend on bystander activation. [Nat Immunol] Mammalian Target of Rapamycin Protein Complex 2 Regulates Differentiation of Th1 and Th2 Cell Subsets Via Distinct Signaling Pathways Researchers showed that conditional deletion of rictor, an essential subunit of mTORC2, impaired differentiation into T helper 1 and T helper 2 cells without diversion into FoxP3+ status or substantial effect on Th17 cell differentiation. [Immunity] Monocytic Suppressive Cells Mediate Cardiovascular Transplantation Tolerance in Mice The results suggest that mobilization of bone marrow CD11b+CD115+Gr1+ monocytes under sterile inflammatory conditions mediates the induction of indefinite allograft survival. [J Clin Invest] PD-L1 has Distinct Functions in Hematopoietic and Nonhematopoietic Cells in Regulating T Cell Responses During Chronic Infection in Mice Here researchers used bone marrow chimeric mice to examine the effects of PD-L1 deficiency in hematopoietic or nonhematopoietic cells during lymphocytic choriomeningitis virus clone 13 (LCMV CL-13) infection. [J Clin Invest] The Endocytic Adaptor Numb Regulates Thymus Size by Modulating Pre-TCR Signaling During Asymmetric Division Researchers found that Numb determines the levels of pre-TCR signaling in dividing thymocytes and, ultimately, the size of the pool from which mature T lymphocytes are selected. [Blood] Notch 1 as a Potential Therapeutic Target in Cutaneous T Cell Lymphoma By immunohistochemistry of 40 biopsies taken from skin lesions of mycosis fungoides and Sezary syndrome researchers demonstrated prominent expression of Notch1 on tumor cells, especially in the more advanced stages. [Blood] Platelet Transcriptional Profile and Protein Expression in Patients with Systemic Lupus Erythematosus: Up-Regulation of the Type I Interferon System is Strongly Associated with Vascular Disease Researchers used microarray to probe the platelet transcriptome in individuals with systemic lupus erythematosus and healthy controls, and the gene and protein expression of a subset of differentially expressed genes was further investigated and correlated to platelet activation status. [Blood] Thymic Stromal Lymphopoietin from Trophoblasts Induces Dendritic Cell-Mediated Regulatory TH2 Bias in the Decidua During Early Gestation in Humans In this study researchers found that human trophoblasts and decidual epithelial cells in maternal-fetal interface of early placentas express thymic stromal lymphopoietins mRNA and protein, but only trophoblast cells secret soluble TSLP. [Blood] Human Fibroblasts Support the Expansion of IL-17 Producing T Cells Via Up-Regulation of IL-23 Production by Dendritic Cells Since researchers have shown that dendritic cells (DCs) interact with fibroblasts during immune responses they studied the impact of skin fibroblasts on human monocyte-derived DC function and subsequent human T cell differentiation. [Blood] Nuclear Alternate Estrogen Receptor GPR30 Mediates 17Beta-Estradiol-Induced Gene Expression and Migration in Breast Cancer-Associated Fibroblasts In this report, researchers show that cancer-associated fibroblasts respond to physiologic concentrations of 17beta-estradiol (E2) by rapidly inducing extracellular signal-regulated kinase phosphorylation and immediate early gene expression, including c-fos and connective tissue growth factor, and cyclin D1. POLICY NEWS NICE Final Draft Guidance Recommends Improved Access to Psoriatic Arthritis Treatments Etanercept (Enbrel), infliximab (Remicade) and adalimumab (Humira) are proposed for treating adults with active and progressive psoriatic arthritis when specified criteria are met. [National Institute of Health and Clinical Excellence, United Kingdom] The Path to Personalized Medicine The leaders of the National Institutes of Health and the Food and Drug Administration have a shared vision of personalized medicine and the scientific and regulatory structure needed to support its growth. Together, they have been focusing on the best ways to develop new therapies and optimize prescribing by steering patients to the right drug at the right dose at the right time. [Food and Drug Administration and the National Institutes of Health, United States] FDA Takes Issue with Genetic Tests from 23andMe, Four Other Firms The Food and Drug Administration has put five genetic test makers on notice that they must get federal approval before marketing their products for use by consumers. [Food and Drug Administration, United States] Diseased Cells Fail to Win Approval In the latest obstacle to expanding the number of embryonic stem-cell lines eligible for US federal funding, a National Institutes of Health (NIH) committee has unanimously rejected dozens of cell lines carrying mutations for specific diseases. [National Institutes of Health, United States] NIH Researchers Explore How Healthy, Young Adults View the Role Genetics Plays in Improving Health Most healthy young adults place greater emphasis on health habits than on genetic risk factors when considering what causes common diseases, a research team from the National Human Genome Research Institute and the Henry Ford Health System in Detroit has found.
