SCIENCE NEWS Mice Engrafted with Human Immune Cells May Provide Clues to Better Prevention and Treatment of Typhoid Fever Better treatments and prevention for typhoid fever may emerge from a laboratory model that has just been developed for the disease. The model is based on transplanting human immune stem cells from umbilical cord blood into mice that are susceptible to infections. [Press release from the University of Washington discussing online prepublication in Proceedings of the National Academy of Sciences USA] Bacteria Identified That May Lead to Inflammatory Bowel Disease in Genetically Susceptible Individuals Certain bacteria that inhabit the intestine provide the environmental trigger that initiates and perpetuates chronic intestinal inflammation in individuals who are genetically susceptible to inflammatory bowel disease, a study has found. [Press release from Harvard School of Public Health discussing online prepublication in Cell Host & Microbe] Discovery Highlights Promise of New Immune System-Based Therapies A new focus on the immune system’s ability to both unleash and restrain its attack on disease has led scientists to identify cells in mice that prevent the immune system from attacking the animals’ own cells, protecting them from autoimmune diseases such as multiple sclerosis, type I diabetes, and lupus. [Press release from the Dana-Farber Cancer Institute discussing online prepublication in Nature]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) Inhibition of Follicular T Helper Cells by CD8+ Regulatory T Cells is Essential for Self Tolerance Here, researchers delineate a CD8+ regulatory T cell lineage that is essential for the maintenance of self tolerance and prevention of murine autoimmune disease. [Nature] The S1P1-mTOR Axis Directs the Reciprocal Differentiation of TH1 and Treg Cells Researchers found that the differentiation of proinflammatory T helper type 1 cells and anti-inflammatory Foxp3+ regulatory T cells was reciprocally regulated by S1P1, a receptor for the bioactive lipid sphingosine 1-phosphate. [Nat Immunol] The Transcription Factor PU.1 Controls Dendritic Cell Development and Flt3 Cytokine Receptor Expression in a Dose-Dependent Manner Here, researchers showed that the transcription factor PU.1 (encoded by Sfpi1) was essential for dendritic cell (DC) development in vivo and that conditional ablation of PU.1 in defined precursors, including the common DC progenitor, blocked Flt3 ligand-induced DC generation in vitro. [Immunity] Function of miR-146a in Controlling Treg Cell-Mediated Regulation of Th1 Responses Here, researchers report that miR-146a, one of the miRNAs prevalently expressed in regulatory T cells, is critical for their suppressor function. [Immunity] Scid IL2r-Gamma-Null Mice are Susceptible to Lethal Salmonella Typhi Infection”>Humanized Nonobese Diabetic-scid IL2r-Gamma-Null Mice are Susceptible to Lethal Salmonella Typhi Infection Here, researchers show that S. Typhi can replicate in vivo in nonobese diabetic-scid IL2r-gamma-null mice engrafted with human hematopoietic stem cells (hu-SRC-SCID mice) to cause a lethal infection with pathological and inflammatory cytokine responses resembling human typhoid. [Proc Natl Acad Sci USA] Promoting Tolerance to Proteolipid Protein-Induced Experimental Autoimmune Encephalomyelitis through Targeting Dendritic Cells Here, researchers report that anti-DEC205-mediated delivery of the self-peptide proteolipid protein (PLP)139-151 to dendritic cells ameliorated clinical symptoms in the PLP-induced SJL model of experimental autoimmune encephalomyelitis. [Proc Natl Acad Sci USA] Cutting Edge: NLRP12 Controls Dendritic and Myeloid Cell Migration to Affect Contact Hypersensitivity Researchers found that murine Nlrp12 , a nucleotide-binding domain leucine-rich repeat linked to atopic dermatitis and hereditary periodic fever in humans, is prominently expressed in dendritic cells and neutrophils. [J Immunol] Soluble CD93 Induces Differentiation of Monocytes and Enhances TLR Responses In the current study, researchers analyzed the functional effects of a soluble form of CD93 on THP-1 monocytic cells and human primary monocytes.
