New Class of ‘Dancing’ Dendritic Cells Derived from Blood Monocytes  ShareThis
Dendritic cells are still notoriously difficult to study in humans. Samples, which come primarily from bone marrow or lymphoid tissue, are simply too difficult to obtain. But new research has shown scientists a way to study “authentic” dendritic cells from mouse monocytes, which are abundant in the blood, a much more accessible source in humans. SCIENCE NEWS

Epratuzumab Reduced Disease Activity in Adult Patients with Moderate to Severe Active Systemic Lupus Erythematosus (SLE)
UCB and U.S. based partner Immunomedics Inc. announced that results from the phase IIB study, EMBLEM(TM), showed that certain doses of epratuzumab were associated with a meaningful and statistically significant reduction in disease activity in adult patients with moderate to severe active SLE. [Press release from UCB discussing data presented at the 74th Annual Scientific Meeting of the American College of Rheumatology in Atlanta]

    Discovery of an Anti-Inflammatory Substance
Biochemists at Kiel University, along with international scientists, have identified an important messenger in the immune system. [Press release from Kiel University discussing online prepublication in Nature Immunology]

Joint Pain Linked to Breast Cancer Drug Not Inflammatory Arthritis or Autoimmune Disease
A new study suggests joint complaints attributed to aromatase inhibitors, popular breast cancer drugs, are not associated with inflammatory arthritis or autoimmune disease. [Press release from Georgetown University Medical Center discussing data presented at the 74th Annual Scientific Meeting of the American College of Rheumatology in Atlanta]

Tiny Variants in a Protein that Alerts the Immune System to the Presence of Infection May Underlie the Rare Ability of Some Individuals to Control HIV Infection Without the Need for Medications
An international research team found that differences in five amino acids in HLA-B are associated with whether or not HIV-infected individuals can control viral levels with their immune system only. [Press release from Massachusetts General Hospital discussing online prepublication in Science]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal)

Microbial Stimulation Fully Differentiates Monocytes to DC-SIGN/CD209+ Dendritic Cells for Immune T Cell Areas
Dendritic cells (DCs), critical antigen-presenting cells for immune control, normally derive from bone marrow precursors distinct from monocytes. It is not yet established if the large reservoir of monocytes can develop into cells with critical features of DCs in vivo . Researchers now show that fully differentiated monocyte-derived DCs develop in mice and DC-SIGN/CD209a marks the cells. [Cell]

The Major Genetic Determinants of HIV-1 Control Affect HLA Class I Peptide Presentation
To define host genetic effects on the outcome of a chronic viral infection, researchers performed genome-wide association analysis in a multiethnic cohort of HIV-1 controllers and progressors, and analyzed the effects of individual amino acids within the classical HLA proteins. [Science]

A Role for IL-27p28 as an Antagonist of gp130-Mediated Signaling
Here, researchers show that IL-27 subunits p28 (IL-27p28), independently of EBI3, antagonized cytokine signaling through gp130 and IL-6-mediated production of IL-17 and IL-10. [Nat Immunol]

Immunosuppressive CD14(+)HLA-DR(low/neg) Monocytes in B Cell Non-Hodgkin’s Lymphoma
Researchers examined the immunophenotype of 40 non-Hodgkin’s lymphoma patients and 27 age-matched controls to better understand systemic immune suppression. [Blood]

Ubiquitination Regulates MHC Class II-Peptide Complex Retention and Degradation in Dendritic Cells
Researchers now show that ubiquitination of MHC class II-peptide complex (pMHC-II) by the E3 ubiquitin ligase membrane-associated RING-CH 1 regulates surface expression, intracellular distribution, and survival of pMHC-II in dendritic cells. [Proc Natl Acad Sci USA]

Role of Antigen Persistence and Dose for CD4+ T Cell Exhaustion and Recovery
Here, researchers dissected the behavior of primed CD4+ T cells exposed to persistent antigen using an inducible transgenic mouse system that allowed them to control antigen presentation as the only experimental variable, independent of the persistent inflammation and disease progression that complicate infectious models. [Proc Natl Acad Sci USA]

Increased B7-H1 Expression on Dendritic Cells Correlates with Programmed Death 1 Expression on T Cells in Simian Immunodeficiency Virus-Infected Macaques and May Contribute to T Cell Dysfunction and Disease Progression
Using the rhesus macaque model, researchers show B7-H1 (programmed death [PD]-L1) is expressed on lymphoid and mucosal dendritic cells (DCs) (both myeloid DCs and plasmacytoid DCs), and its expression significantly increases after simian immunodeficiency virus infection. [J Immunol]

Mesenchymal Stem Cells Contribute to the Abnormal Bone Marrow Microenvironment in Patients with Chronic Idiopathic Neutropenia by Overproduction of Transforming Growth Factor-Beta1
In this study, researchers investigated the possible involvement of bone marrow (BM) mesenchymal stem cells (MSCs) in the pathophysiology of chronic idiopathic neutropenia by assessing the frequency and function of BM MSCs in terms of the proliferative/clonogenic characteristics, the differentiation capacity, the potential to produce pro-inflammatory cytokines and the ability to suppress T cell proliferation. [Stem Cells Dev]

Indoleamine 2,3-Dioxygenase and Regulatory Dendritic Cells Contribute to the Allograft Protection Induced by Infusion of Donor-Specific Splenic Stromal Cells
Here, researchers have used both murine skin and cardiac allograft transplantation models to explore whether in vivo alloresponses can be regulated by infusion with donor-derived splenic stromal cells (SSCs) and to investigate the possible mechanisms by which SSCs exert regulatory effects to prevent allograft rejection. [Cell Mol Immunol]
