    Lymphoid Priming in Human Bone Marrow Begins Before Expression of CD10 with Upregulation of L-Selectin
    Expression of the cell-surface antigen CD10 has long been used to define the lymphoid commitment of human cells. Researchers report a unique lymphoid-primed population in human bone marrow that was generated from hematopoietic stem cells before onset of the expression of CD10 and commitment to the B cell lineage.     The BH3-Only Proteins Bim and Puma Cooperate to Impose Deletional Tolerance of Organ-Specific Antigens
    Researchers showed that Puma cooperates with Bim to impose a thymic-deletion checkpoint to peripheral self-antigens and cemented the notion that defects in apoptosis alone are sufficient to cause autoimmune disease. [Immunity] Abstract | Press Release

    γδ T Cells Recognize a Microbial Encoded B Cell Antigen to Initiate a Rapid Antigen-Specific Interleukin-17 Response
    The authors report that a noted B cell antigen, the algae protein phycoerythrin, is a murine and human γδ T cell antigen. Employing this specificity, they demonstrated that antigen recognition activated naive γδ T cells to make interleukin-17 and respond to cytokine signals that perpetuate the immune response.     T-Bet+ Treg Cells Undergo Abortive Th1 Cell Differentiation due to Impaired Expression of IL-12 Receptor β2
    Researchers demonstrated that the development of T-bet+ regulatory T (Treg) cells that potently inhibit T helper 1 (Th1) cell responses was dependent on the transcription factor STAT1 and occurred directly in response to interferon-produced by effector T cells. [Immunity] Abstract

    Hepatic Expression of HCV RNA-Dependent RNA Polymerase Triggers Innate Immune Signaling and Cytokine Production
    Researchers showed that transient expression of Hepatitis C Virus RNA-dependent RNA polymerase, NS5B, in mouse liver and human hepatocytes resulted in production of small RNA species that activate innate immune signaling via TBK1-IRF3 and NF-B and induced cytokine production, including type I interferons and IL-6.     Regulation of Plasmacytoid Dendritic Cell Responses by PIR-B
    Researchers showed that an inhibitory major histocompatibility complex class I receptor, paired immunoglobulin-like receptor (PIR)-B, suppressed Flt3-L-induced plasmacytoid dendritic cell (PDC) differentiation in bone marrow cells, as well as TLR9-mediated type I interferons-α production by PDCs, through the dephosphorylation of STAT1/2.     Activated Mouse CD4+Foxp3 T Cells Facilitate Melanoma Metastasis via Qa-1-Dependent Suppression of NK-Cell Cytotoxicity
    Scientists found that mitogen-activated CD4+Foxp3 T cells exhibited potent suppressor function on NK-cell proliferation and cytotoxicity in vitro, and notably facilitated B16 melanoma metastasis in vivo. [Cell Res]

    A Novel Mechanism for the Autonomous Termination of Pre-B Cell Receptor Expression via Induction of Lysosomal-Associated Protein Transmembrane 5
    Researchers report that the pre-B cell receptor (pre-BCR) induces the prompt down-modulation of its own expression through induction of lysosomal-associated protein transmembrane 5 that promotes the lysosomal transport and degradation of the intracellular pre-BCR pool, and hence limits the supply of pre-BCR to the cell surface.     CD64 Distinguishes Macrophages from Dendritic Cells in the Gut and Reveals the Th1-Inducing Role of Mesenteric Lymph Node Macrophages during Colitis
    Dendritic cells (DCs) and monocyte-derived macrophages (MFs) are key components of intestinal immunity. However, the lack of surface markers differentiating MFs from DCs has hampered understanding of their respective functions. Researchers demonstrated that, using CD64 expression, MFs can be distinguished from DCs in the intestine of both mice and humans. [Eur J Immunol] Abstract

    Immune Tolerance to Tumor Antigens Occurs in a Specialized Environment of the Spleen
    Researchers found that CD11b+Gr-1intLy6Chi cells expand in the marginal zone of the spleen. Here, they alter the normal tissue cytoarchitecture and closely associate with memory CD8+ T cells, cross-presenting tumor antigens and causing their tolerization. [Cell Rep] Abstract
