PUBLICATIONS(Ranked by impact factor of the journal)Scientists showed that mice with germline deletion of type 2 phosphatidylinositol-5-phosphate 4-kinase gamma (Pip4k2c), the gene encoding PI5P4Kγ, appear normal in regard to growth and viability but have increased inflammation and T-cell activation as they age. Pip4k2c−/− mice had an increase in T-helper-cell populations and a decrease in regulatory T-cell populations with increased proliferation of T cells. [Proc Natl Acad Sci USA] Abstract The authors investigated the phenotypic and functional differences among in vitro activated CD8+ T cells of three different sources, namely naïve, memory and tumor-infiltrating lymphocytes from human and mice, to better understand mechanisms behind potent effector functions and potential for overcoming current limitations. [Sci Rep] Full Article Plasticity of Th17 Cells in Autoimmune Kidney Diseases Investigators analyzed the plasticity of Th17 cells in a mouse model of acute crescentic glomerulonephritis and in a mouse chronic model of lupus nephritis. [J Immunol] Abstract Transgenic expression of Pbx1-d in CD4+ T cells reproduced the phenotypes of Sle1a1 mice, with increased inflammatory functions of CD4+ T cells and impaired Foxp3+ regulatory T cell homeostasis. [J Immunol] Abstract The authors demonstrated that Tpl2 constrains the development of inducible regulatory T cells (iTregs). Tpl2-/- naïve CD4+ T cells preferentially develop into FoxP3+ iTregs in vitro as well as in vivo in a murine model of OVA-induced systemic tolerance. [J Biol Chem] Abstract | Full Article Researchers showed that Irf2bp2 gene expression was decreased in CD4 T cells upon activation. To investigate the possible regulatory roles for IRF2BP2 in CD4 T cell functions, this protein was ectopically expressed in murine primary-activated CD4 T lymphocytes through retroviral transduction. [J Leukoc Biol] Abstract Activation of Myeloid Dendritic Cells, Effector Cells and Regulatory T Cells in Lichen Planus The authors assessed populations of dendritic cells and regulatory and effector T cells in peripheral blood samples collected from patients with lichen planus to evaluate the polyfunctionality of T cells upon toll-like receptor activation. [J Transl Med] Full Article Interleukin (IL)-33 and leflunomide combination therapy resulted in significantly prolonged graft survival, associated with markedly decreased T helper type-1 cells and increased IL-10 levels. In addition, the combination therapy significantly decreased the percentage of CD-45+ B cells on days two and seven, compared with monotherapy. [Lab Invest] Abstract The authors established a novel role of arsenic in regulating immune homeostasis in context to its dose, where, at higher doses, arsenic related upregulation of NF-κB cascade takes on an alternative role that is correlated with increased immune-suppression. [Toxicology] Abstract Investigators utilized human neural precursor cells differentiated from a human induced pluripotent cell line via an embryoid body intermediate stage (EB-NPCs). Intraspinal transplantation of EB-NPCs into mice infected with the neurotropic JHM strain of mouse hepatitis virus resulted in decreased accumulation of CD4+ T cells in the central nervous system that was concomitant with reduced demyelination at the site of injection. REVIEWSThe Role of Lymphatic Niches in T Cell Differentiation The authors highlight the cellularity of various niches within the lymph node and emphasize recent evidence suggesting that distinct types of T cell activation and differentiation occur within different immune contexts in lymphoid organs.
