TOP STORYThe Deubiquitinase OTULIN Is an Essential Negative Regulator of Inflammation and Autoimmunity The authors showed that the methionine-1 (M1)-specific deubiquitinase OTULIN is essential for preventing TNF-associated systemic inflammation in humans and mice. PUBLICATIONS(Ranked by impact factor of the journal)Researchers showed that a severe asthma-associated polymorphism in the gene encoding the interleukin (IL)-4 receptor alpha chain promotes conversion of induced regulatory T cells toward a T helper 17 (TH17) cell fate. [Nat Med] Abstract Researchers report that treatment with low-dose recombinant human interleukin (IL)-2 selectively modulated the abundance of regulatory T cells, follicular helper T (TFH) cells and IL-17-producing helper T cells, but not TH1 or TH2 cells, accompanied by marked reductions of disease activity in patients with systemic lupus erythematosus. [Nat Med] Abstract Scientists report that dendritic cell (DC) activation and immunogenicity are regulated by the transcriptional repressor PCGF6. Pcgf6 was rapidly downregulated upon stimulation, and this downregulation was necessary to permit full DC activation. [Cell Rep] Full Article | Graphical Abstract Inhibition of CDK9 as a Therapeutic Strategy for Inflammatory Arthritis Researchers examined the effects of specific oral small-molecule inhibitors of the transcription regulating cyclin-dependent kinase 9 on the development and progression of collagen-induced arthritis. [Sci Rep] Full Article Regulation of B Cell Differentiation by the Ubiquitin-Binding Protein TAX1BP1 To evaluate the function of Tax1-binding protein 1 (TAX1BP1) in B cells, scientists established TAX1BP1-deficient DT40 B cells that are hyper-responsive to CD40-induced extracellular signal-regulated kinase (ERK) activation signaling, exhibit prolonged and exaggerated ERK phosphorylation and show enhanced B lymphocyte-induced maturation protein 1 expression that is ERK-dependent. [Sci Rep] Full Article Investigators hypothesized that monocyte chemoattractant protein-1-induced protein-1 (MCPIP-1) might play a role in maintaining immune homeostasis in the epithelium both under physiological conditions and upon bacterial infection. [J Innate Immun] Abstract Investigators showed that CD5 expression activates a number of important signaling pathways, including Erk1/2, leading to IL-10 production through a novel pathway independent of B-cell receptor engagement. [Cell Mol Immunol] Abstract Polypyrimidine tract-binding protein (PTBP1) has been implicated in many different processes, however whether PTBP1 plays a broader role in CD4 T cell activation is not known. To examine this question, experiments were designed to introduce shRNA into primary human CD4 T cells to achieve decreased, but not complete ablation of PTBP1 expression. [PLoS One] Full Article Acid Sphingomyelinase (ASM) is a Negative Regulator of Regulatory T Cell (Treg) Development Splenocytes, isolated Naive T lymphocytes and cultured T cells were characterized for various immune T cell markers by flow cytometery. Cell proliferation was measured by Carboxyfluorescein succinimidyl ester dye, cell cycle analysis by propidium iodide, mRNA transcripts by q-RT PCR and protein expression by Western blotting respectively. REVIEWSIgG Subclass Co-Expression Brings Harmony to the Quartet Model of Murine IgG Function A model of murine IgG function is presented in which the co-expression of the IgG subclasses is a central feature, class switching occurs before the commencement of somatic hypermutation, and there is little switching between subclasses. It is named the quartet model to emphasize the harmony that comes from the simultaneous presence of the four subclasses.
