TOP STORYUsing genome-wide dual RNA sequencing of flow-sorted infected and uninfected hepatoma cells the authors showed that the human mucosal immunity gene, mucin-13, was strongly upregulated during Plasmodium exoerythrocytic hepatic-stage infection. [Nat Commun] Full Article | |

PUBLICATIONS(Ranked by impact factor of the journal)Macrophages Target Salmonella by Lc3-Associated Phagocytosis in a Systemic Infection Model Researchers showed that phagocytosis of live but not heat-killed Salmonella triggered recruitment of the autophagy marker GFP-Lc3 in a variety of patterns labeling tight or spacious bacteria-containing compartments, also revealed by electron microscopy. Neutrophils displayed similar GFP-Lc3 associations, but genetic modulation of the neutrophil/macrophage balance and ablation experiments showed that macrophages are critical for the defense response. [Autophagy] Full Article Vaccine Protection against SIVmac239 Acquisition Scientists report significant protection against i.v. acquisition of SIVmac239 using a long-lasting approach to vaccination. The vaccine regimen included a replication-competent herpesvirus engineered to contain a near-full-length SIV genome that expressed all nine SIV gene products, assembled noninfectious SIV virion particles, and was capable of eliciting long-lasting effector-memory cellular immune responses to all nine SIV gene products. [Proc Natl Acad Sci USA] Abstract CD4+ T Cells Promote Humoral Immunity and Viral Control during Zika Virus Infection Researchers investigated the CD4+ T cell response to primary intravenous and intravaginal infection with Zika virus (ZIKV). Using the LysMCre+Ifnar1fl/fl C57BL/6 mouse models, they identified six I-Ab-restricted ZIKV epitopes that stimulated CD4+ T cells with a predominantly cytotoxic Th1 phenotype in mice primed with ZIKV. [PLoS Pathog] Full Article | Editorial Investigators found that gastric Interleukin-17 receptor B (IL-17RB) mRNA and protein were decreased in gastric mucosa of both patients and mice infected with H. pylori. [Cell Death Dis] Full Article Using chronologically aged murine models, researchers demonstrated that decreased ATP production was associated with dysregulated mitochondrial complex expression, enhanced oxidative stress, diminished antioxidant responses, and decreased numbers of healthy mitochondria in aged adult macrophages and lung in response to S. pneumoniae. [Sci Rep] Full Article To investigate the effect of age on influenza-specific CD4 T cell specificity and functionality, reactivity in cohorts of two year old children and young adult subjects was compared. [Sci Rep] Full Article To evaluate the potential of immune responses to protect against malaria, the authors expressed four P. knowlesi antigens in rhesus cytomegalovirus 68-1 or in Rh189-deleted 68-1, which additionally elicited canonical MHC-Ia-restricted CD8+ T cells. [PLoS One] Full Article | Press Release High titers of cross-protective, broadly neutralizing antibodies were elicited by a single dose of recombinant vesicular stomatitis virus (rVSV)-severe fever with thrombocytopenia virus (SFTSV)/AH12-GP in both immunocompetent and immunocompromised mice against multiple strains of SFTSV and the related but distinct phlebovirus Heartland virus. REVIEWSImmunometabolism at the Interface between Macrophages and Pathogens It is generally regarded that the progression of an infection within host macrophages is the consequence of a failed immune response. However, recent appreciation of macrophage heterogeneity, with respect to both development and metabolism, indicates that the reality is more complex. [Nat Rev Immunol] Abstract Adaptive Immune Responses to Primary and Secondary Dengue Virus Infections The authors contrast the mechanisms initiating and sustaining adaptive immune responses during primary infection with the immune pathways that are pre-existing and reactivated during secondary dengue. INDUSTRY NEWSTakeda’s Dengue Vaccine Candidate Meets Primary Endpoint in Pivotal Phase III Efficacy Trial Takeda Pharmaceutical Company Limited announced that the pivotal Phase III trial of its dengue vaccine candidate met the primary efficacy endpoint. This first analysis of the Tetravalent Immunization against Dengue Efficacy Study trial showed that the company’s investigational live-attenuated tetravalent dengue vaccine was efficacious in preventing dengue fever caused by any of the four serotypes of the virus. [Takeda Pharmaceutical Company Limited] Press Release NIAID-Sponsored Clinical Trial of Ebola Vaccines Begins at Cincinnati Children’s A Phase I clinical trial of investigational vaccines intended to protect against Zaire ebolavirus (Ebola) is underway at Cincinnati Children’s Hospital Medical Center in the United States. [Cincinnati Children’s Hospital Medical Center] Press Release Merck announced that V114, the company’s investigational 15-valent pneumococcal conjugate vaccine, has received Breakthrough Therapy Designation from the FDA for the prevention of invasive pneumococcal disease caused by the vaccine serotypes in pediatric patients 6 weeks to 18 years of age. POLICY NEWSUS Government Shutdown’s Effects on Science Ripple Overseas From canceled conferences to delayed publications, fallout of the shutdown spread beyond US borders, prompting concerns about long-term damage to international collaboration. [The Scientist] Editorial How Brexit Threatens Irish Science’s Cross-Border Collaboration Scientists in Northern Ireland and the Republic of Ireland are worried that Brexit will upset the island’s research ecosystem, which has benefited from peace and economic growth over the past 20 years.
