    TOP STORY
Intestinal Inhibition of Atg7 Prevents Tumor Initiation through a Microbiome-Influenced Immune Response and Suppresses Tumour Growth
Researchers showed that autophagy is activated in the intestinal epithelium in murine and human colorectal cancer and that the conditional inactivation of Atg7 in intestinal epithelial cells inhibits the formation of pre-cancerous lesions in Apc+/− mice by enhancing anti-tumor responses. [Nat Cell Biol] Abstract

    INTESTINAL CANCERS & DISEASES

Preventing E-Cadherin Aberrant N-Glycosylation at Asn-554 Improves Its Critical Function in Gastric Cancer
Using transgenic mice models and human clinical samples, researchers demonstrated that N-acetylglucosaminyltransferase V-mediated glycosylation caused an abnormal pattern of E-cadherin expression in the gastric mucosa. [Oncogene] Abstract

PAK5-Mediated E47 Phosphorylation Promotes Epithelial–Mesenchymal Transition and Metastasis of Colon Cancer
Scientists found that p21-activated kinase 5 (PAK5)-mediated E47 phosphorylation promoted epithelial–mesenchymal transition in advanced colon cancer. PAK5 interacted with E47 and phosphorylated E47 on Ser39 under hepatocyte growth factor stimulation, which decreased cell–cell cohesion, increased cell migration and invasion in vitro and promoted metastasis in a xenograft model. [Oncogene] Abstract

Tight Junction CLDN2 Gene Is a Direct Target of the Vitamin D Receptor
Using whole body VDR-/- mice, intestinal epithelial VDR conditional knockout (VDRΔIEC) mice, and cultured human intestinal epithelial cells, investigators demonstrated that the CLDN2 gene is a direct target of the transcription factor VDR. [Sci Rep] Full Article

IL-18 Is Involved in Eosinophil-Mediated Tumoricidal Activity against a Colon Carcinoma Cell Line by Upregulating LFA-1 and ICAM-1
Given the role of IL-18 in cell adhesion and in protecting against colon cancer, the authors evaluated its role in eosinophil-mediated cytotoxicity against Colo-205, a human colon carcinoma cell line. [J Immunol] Abstract

Prognostic Relevance of LGALS3BP in Human Colorectal Carcinoma
The role of LGALS3BP in colorectal cancer (CRC) prognosis was investigated. They provided evidence that low expression of LGALS3BP participates in malignant progression of CRC and implicated poor prognosis. [J Transl Med] Full Article

Retinoic Acid Morpholine Amide (RAMA) Inhibits Expression of Fas Ligand through EP1 Receptor in Colon Cancer Cells
Investigators demonstrated that RAMA significantly suppressed FasL expression in colon cancer cells in a dose- and time-dependent manner. [Tumour Biol] Abstract

Distribution of Intestinal Stem Cell Markers in Colorectal Precancerous Lesions
The authors investigated the expression and distribution of intestinal stem cell markers in serrated polyps and conventional adenomas. [Histopathology] Abstract

INTESTINAL STEM CELL & ORGANOID RESEARCH

Small Molecule and RNAi Induced Phenotype Transition of Expanded and Primary Colonic Epithelial Cells
Scientists report primary and prolonged culture of normal porcine epithelial cells from colon for cell line development. In addition, a novel primary three-dimensional intestinal culture system was presented, which generated organoids composed of a highly polarized epithelial layer lining a core of subepithelial tissue. REVIEWS
Epigenetic Regulation of the Intestinal Epithelium
The authors describe the role that epigenetics plays in intestinal homeostasis and disease, with an emphasis on results from mouse models. They highlight the importance of producing and analyzing next-generation sequencing data detailing the epigenome from intestinal stem cell to differentiated intestinal villus cell. [Cell Mol Life Sci] Abstract
