    Tuft-Cell-Derived IL-25 Regulates an Intestinal ILC2–Epithelial Response Circuit
    Scientists showed that tuft cells constitutively express interleukin (IL)-25 to sustain tissue-resident group 2 innate lymphoid cells (ILC2) homeostasis in the resting lamina propria in mice. After helminth infection, tuft-cell-derived IL-25 further activated ILC2s to secrete IL-13, which acts on epithelial crypt progenitors to promote differentiation of tuft and goblet cells, leading to increased frequencies of both.     Contributions of Microbiome and Mechanical Deformation to Intestinal Bacterial Overgrowth and Inflammation in a Human Gut-on-a-Chip
    A human gut-on-a-chip microdevice was used to coculture multiple commensal microbes in contact with living human intestinal epithelial cells for more than a week in vitro and to analyze how gut microbiome, inflammatory cells, and peristalsis-associated mechanical deformations independently contribute to intestinal bacterial overgrowth and inflammation.     Network-Level Effects of Kinase Inhibitors Modulate TNF-α-Induced Apoptosis in the Intestinal Epithelium
    Researchers found that inhibitors of the mitogen-activated protein kinase kinase kinase Raf, MEK, or extracellular signal-regulated kinase exerted distinct effects on the timing and magnitude of tumor necrosis factor-α (TNF-α)-induced apoptosis in the mouse intestine. [Sci Signal] Abstract

    Treating Colon Cancer Cells with FK228 Reveals a Link between Histone Lysine Acetylation and Extensive Changes in the Cellular Proteome
    The authors used stable isotope labeling by amino acids in cell culture and affinity enrichment followed by high-resolution liquid chromatograph-mass spectrometer (LC-MS)/MS analysis to quantitate the changes in the lysine acetylome in HCT-8 cells after FK228 treatment. [Sci Rep] Full Article
