How Breast Tissue Stiffening Promotes Breast Cancer Development

Researchers provide new insight into how the stiffening of breast tissue plays a role in breast cancer development. By examining how mammary cells respond in a stiffness-changing hydrogel, they discovered that several pathways work together to promote the transformation of breast cells into cancer cells. Signaling Pathway of Globo-Series Glycosphingolipids and β1,3-Galactosyltransferase V (β3GalT5) in Breast Cancer

Investigators showed that the expression of β3GalT5 significantly correlated with tumor progression and poor survival in patients, and the globo-series glycosphingolipids in breast cancer cells form a complex in membrane lipid raft with caveolin-1 and focal adhesion kinase which then interact with AKT and receptor-interacting protein kinase, respectively. Thymidylate Synthase Maintains the De-Differentiated State of Triple Negative Breast Cancers

SPAG5 Upregulation Contributes to Enhanced c-MYC Transcriptional Activity via Interaction with c-MYC Binding Protein in Triple-Negative Breast Cancer

Sperm-associated antigen 5 (SPAG5)-silenced cells were more sensitive to the PARP inhibitor olaparib. Mechanistically, SPAG5 interacted with c-MYC binding protein (MYCBP), thereby increasing MYCBP protein levels and leading to increased c-MYC transcriptional activity, which promoted the expression of the c-MYC target genes: CDC20, CDC25C, BRCA1, BRCA2, and RAD51. CFP Suppresses Breast Cancer Cell Growth by TES-Mediated Upregulation of the Transcription Factor DDIT3

Glucocorticoid Receptor-IRS-1 Axis Controls EMT and the Metastasis of Breast Cancers

Scientists investigated the role of glucocorticoid receptor (GR) and its underlying mechanism in mediating breast cancer cell survival and metastasis. Plasminogen Activator Inhibitor 1 (PAI1) Promotes Actin Cytoskeleton Reorganization and Glycolytic Metabolism in Triple Negative Breast Cancer

Researchers showed that increased PAI1 drove rearrangement of the actin cytoskeleton, mitochondrial fragmentation, and glycolytic metabolism in triple negative breast cancer cells. In two-dimensional environments, both stable expression of PAI1 and treatment with recombinant PAI1 increased migration, which could be blocked with the specific inhibitor tiplaxtinin. MicroRNA-708 Activation by Glucocorticoid Receptor Agonists Regulate Breast Cancer Tumorigenesis and Metastasis via Downregulation of NF-κB Signaling

Investigators found that treatment with either the synthetic glucocorticoid dexamethasone or the natural glucocorticoid mimic, antcin A significantly increased miR-708 expression by transactivation of glucocorticoid receptor alpha in MCF-7 and MDA-MB-231 human breast cancer cells. AKT‐Dependent Sugar Addiction by Benzyl Isothiocyanate in Breast Cancer Cells

Cellular studies using MDA‐MB‐231 and SUM159 human breast cancer cell lines revealed benzyl isothiocyanate-mediated induction and punctate localization of glucose transporter GLUT‐1, which was accompanied by an increase in intracellular pyruvate levels. [Mol Carcinog] Abstract

In Vitro Cytotoxicity and Anticancer Effects of Citral Nanostructured Lipid Carrier on MDA MBA-231 Human Breast Cancer Cells

Cytotoxicity of Eupatorin in MCF-7 and MDA-MB-231 Human Breast Cancer Cells via Cell Cycle Arrest, Anti-Angiogenesis and Induction of Apoptosis

Eupatorin’s effect on the human breast carcinoma cell line MCF-7 and MDA-MB-231 was investigated. MTT assay showed that eupatorin had cytotoxic effects on MCF-7 and MDA-MB-231 cells but was non-toxic to the normal cells of MCF-10a in a time-dose dependent manner. [Sci Rep] Full Article

Draft Federal Spending Deal Includes Largest Boost for FDA in Five Years

A government funding deal released includes $269 million in new funding for the FDA, which would represent the largest increase the agency has seen in at least five years. With that new funding, the FDA would receive $3.08 billion in government funding for fiscal year 2019. Coupled with roughly $2.5 billion in fees from drug, device, and other regulated companies, FDA’s total budget would be $5.67 billion. [STAT News] Editorial
