O-Linked α2,3 Sialylation Defines Stem Cell Populations in Breast Cancer

Scientists identified a lectin, SLBR-N, which bound to O-linked α2,3 sialic acids, that was able to enrich for cancer stem cells (CSCs) in vitro and in vivo. This O-glycan was expressed on CD44 and promoted its interaction with hyaluronic acid, facilitating CD44 signaling and CSC properties.

MAPK4 Promotes Triple Negative Breast Cancer Growth and Reduces Tumor Sensitivity to PI3K Blockade

Researchers demonstrated that a large subset of TNBC expressed significant levels of MAPK4, and this expression was critical for driving AKT activation independent of PI3K and promoting TNBC cell and xenograft growth.

C/EBPB-Dependent Adaptation to Palmitic Acid Promotes Tumor Formation in Hormone Receptor Negative Breast Cancer

Investigators demonstrated that obesity-associated tumor formation was driven by cellular adaptation rather than expansion of pre-existing clones within the cancer cell population.

Exosomal circCARM1 from Spheroids Reprograms Cell Metabolism by Regulating PFKFB2 in Breast Cancer

The authors investigated breast cancer cell metabolism reprogramming by circular RNAs (circRNAs) from breast cancer stem cell (BCSC) exosomes and found that BCSC exosome-derived circCARM1 played an important role in breast cancer cell glycolysis by sponging miR-1252-5p.

    Abstract

RNA-Binding Protein p54nrb/NONO Potentiates Nuclear EGFR-Mediated Tumorigenesis of Triple-Negative Breast Cancer

Scientists indicated that NONO enhanced nuclear epidermal growth factor receptor (EGFR)-mediated tumorigenesis and may be a potential therapeutic target for TNBC patients with nuclear EGFR expression.

Targeting Prooxidant MnSOD Effect Inhibits Triple-Negative Breast Cancer (TNBC) Progression and M2 Macrophage Functions under the Oncogenic Stress

Knockdown of MnSOD inhibited TNBC cell invasion, breast cancer stem cells, mitochondrial reactive oxygen species, and interleukin-6 excretion promoted by multiple copies in T cell malignancy-1.

Endoplasmic Reticulum Stress Inhibits AR Expression via the PERK/eIF2α/ATF4 Pathway in Luminal Androgen Receptor Triple-Negative Breast Cancer and Prostate Cancer

Investigators demonstrated that endoplasmic reticulum stress could decrease androgen receptor mRNA and protein levels via PERK/eIF2α/ATF4 signaling in luminal androgen receptor TNBC and prostate cancer.

Stem-Like Breast Cancer Cells in the Activated State Resist Genetic Stress via TGFBI-ZEB1

The authors characterized a population of stem-like breast cancer cells expressing the integrin αvβ3 as transcriptionally related to activated stem/basal cells in the normal human mammary gland.

KDM6A Depletion in Breast Epithelial Cells Leads to Reduced Sensitivity to Anticancer Agents and Increased TGF-β Activity

Using MCF10A as a model of non-tumorigenic epithelial breast cells, scientists found that silencing KDM6A promoted cell migration and transformation demonstrated by the formation of tumor-like acini in 3D culture.

    Abstract

LncRNA UCA1 Promotes SOX12 Expression in Breast Cancer by Regulating m6A Modification of miR-375 by METTL14 through DNA Methylation

Researchers elucidated the underlying mechanism by which lncRNA UCA1 affected the m6A modification of miR-375 by mediating the DNA methylation of METTL14 and then altering SOX12 expression in breast cancer.
