    SCIENCE NEWS

How Does Estrogen Feed Breast Cancer Tumors?
Researchers found that estrogen inhibits a protein called MLK3 that causes normal cell death. Blocking MLK3 leads to uncontrolled growth of cancer cells and resistance to chemotherapy. [Press release from Loyola University discussing online prepublication in Cancer Research]

Gene Test Hope for Personalised Breast Cancer Treatment
Scientists have developed a system to identify faulty or missing genes that could prevent specific chemotherapy regimes from working. [Press release from Cancer Research UK discussing online prepublication in Lancet Oncology]

PINC Trial Launched to Test New Treatment for Pre-Invasive Breast Cancer
This three-year clinical trial will test the effectiveness of the anti-malarial drug chloroquine in treating 90 women with ductal carcinoma in situ (DCIS), a type of breast cancer in which the cancer cells start in the milk ducts but have not yet become invasive and spread in the breast. [George Mason University Press Release]

Freezing Breast Tumors Helps Stop Cancer’s Spread in Mice, University Of Michigan Study Finds
Freezing a cancer kills it in its place, and also appears to generate an immune response that helps stop the cancer’s spread, leading to improved survival rates over surgery. [Press release from University Of Michigan discussing online prepublication in Annals of Surgical Oncology]

Researchers aimed to establish agreement between microarray-based single sample predictors for identification of breast cancer molecular subtypes. [Lancet Oncol]

LRP6 Overexpression Defines a Class of Breast Cancer Subtype and is a Target for Therapy
Studies highlight LRP6 as a potential therapeutic target in breast cancer, and introduce Mesd as a promising antitumor agent for treating breast cancer subtypes with Wnt activation at the cell surface. [Proc Natl Acad Sci USA]

Dose-Dependent Induction of Distinct Phenotypic Responses to Notch Pathway Activation in Mammary Epithelial Cells
Researchers investigated the cellular basis for the heterogeneous responses to Notch pathway activation in 3D cultures of MCF-10A mammary epithelial cells. [Proc Natl Acad Sci USA]

Disruption of Laminin-Integrin-CD151-Focal Adhesion Kinase Axis Sensitizes Breast Cancer Cells to ErbB2 Antagonists
Adhesion of human ErbB2+ breast cancer cells to basement membrane laminin-5 provides substantial resistance to trastuzumab and lapatinib, agents that respectively target the extracellular and kinase domains of ErbB2. [Cancer Res]

Downregulation of Notch Pathway by a Gamma-Secretase Inhibitor Attenuates AKT/Mammalian Target of Rapamycin Signaling and Glucose Uptake in an ERBB2 Transgenic Breast Cancer Model
In this study, researchers use an ERBB2-transgenic mouse model of breast cancer (neuT) to show that Notch signaling plays a critical role in tumor maintenance. [Cancer Res]

Nutrient Sensor O-GlcNAc Transferase Regulates Breast Cancer Tumorigenesis Through Targeting of the Oncogenic Transcription Factor FoxM1
Findings identify O-linked beta-N-acetylglucosamine (O-GlcNAc) as a novel mechanism through which alterations in glucose metabolism regulate cancer growth and invasion and suggest that O-GlcNAc transferase may represent novel therapeutic targets for breast cancer. [Oncogene]

    CLINICAL RESEARCH

Comparable Survival Between pN0 Breast Cancer Patients Undergoing Sentinel Node Biopsy and Extensive Axillary Dissection: A Report From the Korean Breast Cancer Society
In this study, the survival and recurrence after sentinel node biopsy alone and axillary lymph node dissection in pN0 Korean breast cancer patients were investigated. [J Clin Oncol]

Breast Cancer Subtypes and the Risk of Local and Regional Relapse
The risk of local and regional relapse associated with each breast cancer molecular subtype was determined in a large cohort of patients with breast cancer. Subtype assignment was accomplished using a validated six-marker immunohistochemical panel applied to tissue microarrays. [J Clin Oncol]

Assessment of an RNA Interference Screen-Derived Mitotic and Ceramide Pathway Metagene as a Predictor of Response to Neoadjuvant Paclitaxel for Primary Triple-Negative Breast Cancer: A Retrospective Analysis of Five Clinical Trials

The paclitaxel response metagene shows promise as a paclitaxel-specific predictor of pathological complete response in patients with triple-negative breast cancer. [Lancet Oncol]

Plasma Homocysteine and Cysteine and Risk of Breast Cancer in Women
Although this study offers little support for an association between circulating homocysteine and overall breast cancer risk, higher homocysteine levels may be associated with an increased risk for breast cancer among women with low folate status. The increased risk of breast cancer associated with high cysteine levels warrants further investigation. [Cancer Res]

Intraoperative Imprint Cytology Examination of Sentinel Lymph Nodes After Neoadjuvant Chemotherapy in Breast Cancer Patients
Researchers evaluated the validity of intraoperative imprint cytology for sentinel lymph nodes testing in patients after neoadjuvant chemotherapy. [Ann Surg Oncol]
