TOP STORY Discovery Halts Breast Cancer Stem Cells Breast cancer stem cells (CSCs) are stimulated by estrogen via a pathway that mirrors normal stem cell development. Disrupting the pathway, researchers were able to halt the expansion of breast CSCs, a finding that suggests a new drug therapy target. CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) LABORATORY RESEARCH The ER UDPase ENTPD5 Promotes Protein N-Glycosylation, the Warburg Effect, and Proliferation in the PTEN Pathway PI3K and PTEN lipid phosphatase control the level of cellular phosphatidylinositol (3,4,5)-trisphosphate, an activator of AKT kinases that promotes cell growth and survival. Mutations activating AKT are commonly observed in human cancers. Researchers report here that ENTPD5, an endoplasmic reticulum enzyme, is upregulated in cell lines and primary human tumor samples with active AKT. [Cell] STAT3-Induced S1PR1 Expression is Crucial for Persistent STAT3 Activation in Tumors Researchers show that expression of sphingosine-1-phosphate receptor-1, a G protein-coupled receptor for the lysophospholipid sphingosine-1-phosphate, is elevated in STAT3-positive tumors. [Nat Med] Specific Posttranslational Modification Regulates Early Events in Mammary Carcinoma Formation To determine the molecular basis of how early events in breast carcinoma formation are regulated by GnT-V, researchers studied both the early stages of mammary tumor formation by using 3D cell culture and a her-2 transgenic mouse mammary tumor model. [Proc Natl Acad Sci USA] Estrogen Expands Breast Cancer Stem-Like Cells through Paracrine FGF/Tbx3 Signaling Here, researchers report that, in analogy to embryonic mammary epithelial biology, estrogen signaling expands the pool of functional breast cancer stem cells through a paracrine FGF/FGFR/Tbx3 signaling pathway. [Proc Natl Acad Sci USA] Astrocyte Elevated Gene-1 (AEG-1) Functions as an Oncogene and Regulates Angiogenesis Researchers provide definitive evidence that astrocyte-elevated gene-1 (AEG-1) is indeed a transforming oncogene and show that stable expression of AEG-1 in normal immortal cloned rat embryo fibroblast cells induces morphological transformation and enhances invasion and anchorage-independent growth in soft agar, two fundamental biological events associated with cellular transformation. [Proc Natl Acad Sci USA] HER2 Silences Tumor Suppression in Breast Cancer Cells by Switching Expression of C/EBPBeta Isoforms In this study, researchers show that HER2-overexpressing breast cancer cells avert TGFbeta- and oncogene-induced senescence-mediated tumor suppression by switching expression of two functionally distinct isoforms of the transcription factor C/EBPbeta, which has been implicated previously in breast cancer development. [Cancer Res] Epigenetically Deregulated microRNA-375 Is Involved in a Positive Feedback Loop with Estrogen Receptor Alpha in Breast Cancer Cells Estrogen receptor alpha (ERalpha) upregulation causes abnormal cell proliferation in about two thirds of breast cancers, yet understanding of the underlying mechanisms remains incomplete. Here, researchers show that high expression of the microRNA miR-375 in ERalpha-positive breast cell lines is a key driver of their proliferation. [Cancer Res] Effects of Human Mesenchymal Stem Cells on ER-Positive Human Breast Carcinoma Cells Mediated through ER-SDF-1/CXCR4 Crosstalk Adult human mesenchymal stem cells (hMSCs) have been shown to home to sites of carcinoma and affect biological processes, including tumor growth and metastasis. Previous findings have been conflicting and a clear understanding of the effects of hMSCs on cancer remains to be established. Therefore, researchers set out to investigate the impact of hMSCs on the estrogen receptor positive, hormone-dependent breast carcinoma cell line MCF-7. [Mol Cancer] CLINICAL RESEARCH Using Mammographic Density to Predict Breast Cancer Risk: Dense Area or Percent Dense Area Researchers measured mammographic density, including non-dense area (here a surrogate for weight), in the medio-lateral oblique (MLO) mammogram using a computer-assisted thresholding technique for 634 cases and 1880 age-matched controls from the Cambridge and Norwich Breast Screening Programs.
