Fibronectin Conformation Regulates the Proangiogenic Capability of Tumor-Associated Adipogenic Stromal Cells
The specific effect of fibronectin (Fn) conformation on tumor angiogenesis is undefined. The findings suggest that tumor-associated partial unfolding of Fn decreases adhesion while enhancing vascular endothelial growth factor secretion by breast cancer-associated adipogenic precursor cells, and that altered integrin specificity may underlie these changes. WWP1 E3 Ligase Targets LATS1 for Ubiquitin-Mediated Degradation in Breast Cancer Cells
Investigators have provided in vitro and in vivo evidence that WWP1 is essential for Large Tumor Suppressor 1 (LATS1) stability and negatively regulate LATS1 by promoting LATS1 degradation through polyubiquitination and the 26S proteasome pathway. Importantly, they also showed that degradation of LATS1 is critical in mediating WWP1-induced increased cell proliferation in breast cancer cells. [PLoS One] Full Article

Non-Invasive Analysis of Acquired Resistance to Cancer Therapy by Sequencing of Plasma DNA
Researchers reported sequencing of cancer exomes in serial plasma samples to track genomic evolution of metastatic cancers in response to therapy. Six patients with advanced breast, ovarian and lung cancers were followed over 1-2 years. [Nature] Abstract

Alcohol Consumption Before and After Breast Cancer Diagnosis: Associations with Survival from Breast Cancer, Cardiovascular Disease, and Other Causes
Overall alcohol consumption before diagnosis was not associated with disease-specific survival, but researchers found a suggestion favoring moderate consumption. There was no evidence for an association with postdiagnosis alcohol intake and breast cancer survival. This study, however, does provide support for a benefit of limited alcohol intake for cardiovascular and overall survival in women with breast cancer. [J Clin Oncol] Abstract | Press Release

Adjuvant Endocrine Therapy Initiation and Persistence in a Diverse Sample of Patients with Breast Cancer
Adjuvant endocrine therapy for breast cancer reduces recurrence and improves survival rates. Many patients never start treatment or discontinue prematurely. A better understanding of factors associated with endocrine therapy initiation and persistence could inform practitioners how to support patients. Scientists reviewed the available literature data on: the origin of androgens after menopause, the association between circulating androgens and breast cancer (BC) incidence and recurrence, the relationship between circulating and intratumoral hormones, the prognostic significance of the presence of androgen receptors in the different BC subtypes, the androgen effect on BC cell lines, and the relationship between androgens and aromatase inhibitors. Excess Estrogen in Pregnancy Can Silence BRCA1 in Daughters, Increasing Breast Cancer Risk
Excess estrogen levels during pregnancy can disable, in their daughters, a powerful breast cancer tumor suppressor gene. Researchers found the DNA repair gene BRCA1 to be silenced in one year-old girls exposed to a high hormonal fetal environment. The researchers say their study suggests that BRCA1 silencing by methylation in utero may be an important mechanism that increases breast cancer risk later in life. Nearly Half of Breast Cancer Patients at High Risk of Carrying BRCA Mutations Do Not Receive Genetic Testing Recommendations from Physicians
