    TOP STORY
Signature of Circulating Breast Tumor Cells that Spread to the Brain Found
Some breast tumor circulating cells in the bloodstream are marked by a constellation of biomarkers that identify them as those destined to seed the brain with a deadly spread of cancer, said researchers. [Press release from the Baylor College of Medicine discussing online prepublication in Science Translational Medicine] Press Release | Abstract

    LABORATORY RESEARCH

Boron Neutron Capture Therapy Demonstrated in Mice Bearing EMT6 Tumors following Selective Delivery of Boron by Rationally Designed Liposomes
The application of boron neutron capture therapy following liposomal delivery of a 10B-enriched polyhedral borane and a carborane against mouse mammary adenocarcinoma solid tumors was investigated. [Proc Natl Acad Sci USA] Abstract | Press Release

Novel Role of MicroRNA146b in Promoting Mammary Alveolar Progenitor Cell Maintenance
Researchers have shown that microRNA146b promotes the maintenance of pregnancy derived mammary luminal alveolar progenitors. [J Cell Sci] Abstract

Fibronectin Conformation Regulates the Proangiogenic Capability of Tumor-Associated Adipogenic Stromal Cells
The specific effect of fibronectin (Fn) conformation on tumor angiogenesis is undefined. The findings suggest that tumor-associated partial unfolding of Fn decreases adhesion while enhancing vascular endothelial growth factor secretion by breast cancer-associated adipogenic precursor cells, and that altered integrin specificity may underlie these changes. [Biochim Biophys Acta] Abstract

Integrin Binding Protein Nischarin Interacts with Tumor Suppressor Liver Kinase B1(LKB1) to Regulate Cell Migration of Breast Epithelial Cells
Researchers demonstrated functional interaction between LKB1 and Nischarin to inhibit cell migration and breast tumor progression. [J Biol Chem] Abstract | Full Article

Hyperinsulinemia Promotes Metastasis to the Lung in a Mouse Model of Her2/Neu-Mediated Breast Cancer
Scientists used a mouse model of Her2-mediated breast cancer on a background of hyperinsulinemia to determine how elevated circulating insulin levels affect Her2-mediated primary tumor growth and lung metastasis. [Endocr Relat Cancer] Abstract

WWP1 E3 Ligase Targets LATS1 for Ubiquitin-Mediated Degradation in Breast Cancer Cells
Investigators have provided in vitro and in vivo evidence that WWP1 is essential for Large Tumor Suppressor 1 (LATS1) stability and negatively regulate LATS1 by promoting LATS1 degradation through polyubiquitination and the 26S proteasome pathway. Importantly, they also showed that degradation of LATS1 is critical in mediating WWP1-induced increased cell proliferation in breast cancer cells. [PLoS One] Full Article

Expression and Functional Role of Sprouty-2 in Breast Morphogenesis
Researchers analyzed the expression of Sprouty-2 in the mammary gland and studied its role in branching morphogenesis. [PLoS One] Full Article

    CLINICAL RESEARCH

Non-Invasive Analysis of Acquired Resistance to Cancer Therapy by Sequencing of Plasma DNA
Researchers reported sequencing of cancer exomes in serial plasma samples to track genomic evolution of metastatic cancers in response to therapy. Six patients with advanced breast, ovarian and lung cancers were followed over 1-2 years. [Nature] Abstract

Alcohol Consumption Before and After Breast Cancer Diagnosis: Associations with Survival from Breast Cancer, Cardiovascular Disease, and Other Causes
Overall alcohol consumption before diagnosis was not associated with disease-specific survival, but researchers found a suggestion favoring moderate consumption. There was no evidence for an association with postdiagnosis alcohol intake and breast cancer survival. This study, however, does provide support for a benefit of limited alcohol intake for cardiovascular and overall survival in women with breast cancer. [J Clin Oncol] Abstract | Press Release

Adjuvant Endocrine Therapy Initiation and Persistence in a Diverse Sample of Patients with Breast Cancer
Adjuvant endocrine therapy for breast cancer reduces recurrence and improves survival rates. Many patients never start treatment or discontinue prematurely. A better understanding of factors associated with endocrine therapy initiation and persistence could inform practitioners how to support patients. The authors analyzed data from a longitudinal study of 2,268 women diagnosed with breast cancer. [Breast Cancer Res Treat] Abstract | Press Release

    REVIEWS

Postmenopausal Breast Cancer, Androgens, and Aromatase Inhibitors
Scientists reviewed the available literature data on: the origin of androgens after menopause, the association between circulating androgens and breast cancer (BC) incidence and recurrence, the relationship between circulating and intratumoral hormones, the prognostic significance of the presence of androgen receptors in the different BC subtypes, the androgen effect on BC cell lines, and the relationship between androgens and aromatase inhibitors. [Breast Cancer Res Treat] Abstract

Metabolic Syndrome and Postmenopausal Breast Cancer: Systematic Review and Meta-Analysis
The role of metabolic syndrome (MS) and its individual components in postmenopausal breast cancer (PBC) risk is still unclear. The authors reviewed and summarized epidemiological studies assessing the association of MS with the risk of PBC. [Menopause] Abstract

    SCIENCE NEWS

Excess Estrogen in Pregnancy Can Silence BRCA1 in Daughters, Increasing Breast Cancer Risk
Excess estrogen levels during pregnancy can disable, in their daughters, a powerful breast cancer tumor suppressor gene. Researchers found the DNA repair gene BRCA1 to be silenced in one year-old girls exposed to a high hormonal fetal environment. The researchers say their study suggests that BRCA1 silencing by methylation in utero may be an important mechanism that increases breast cancer risk later in life. [Press release from Georgetown University Medical Center discussing research presented at the American Association of Cancer Research (AACR) Annual Meeting 2013, Washington] Press Release

Nearly Half of Breast Cancer Patients at High Risk of Carrying BRCA Mutations Do Not Receive Genetic Testing Recommendations from Physicians
Only 53 percent of newly diagnosed breast cancer patients who were at high risk of carrying a BRCA 1 or BRCA 2 mutation – based on age, diagnosis, and family history of breast or ovarian cancer – reported that their doctors urged them to be tested for the genes. [Press release from the Perelman School of Medicine at the University of Pennsylvania discussing research presented at the AACR Annual Meeting 2013, Washington] Press Release
