    TOP STORY
Expression and Therapeutic Targeting of Dopamine Receptor-1 (D1R) in Breast Cancer
Dopamine and D1R agonists — signaling through the cGMP/protein kinase G pathway — suppressed cell viability, inhibited invasion and induced apoptosis in multiple breast cancer cell lines. [Oncogene] Abstract

    LABORATORY RESEARCH

Runx2 Contributes to the Regenerative Potential of the Mammary Epithelium
Researchers investigated the role of Runx2 in the mammary stem/progenitor population and its relationship with WNT signaling. They showed that RUNX2 protein is differentially expressed throughout embryonic and adult development of the murine mammary gland with high levels of expression in mammary stem-cell enriched cultures. [Sci Rep] Full Article

MiR-129-3p Promotes Docetaxel Resistance of Breast Cancer Cells via CP110 Inhibition
Scientists investigated the possible role of miR-129-3p in breast cancer cell resistance to docetaxel. MiR-129 and miR-129-3p inhibitor were transfected into breast cancer cells to investigate their effects on chemoresistance to docetaxel. [Sci Rep] Full Article

Androgen Receptor Promotes Tamoxifen Agonist Activity by Activation of EGFR in ERα-Positive Breast Cancer
The authors discovered that androgen receptor is overexpressed in cells with decreased Rho guanine nucleotide dissociation inhibitor (Rho GDIα) and sought to determine AR’s contribution to resistance. They engineered estrogen receptor (ER)α-positive cell lines with stable knockdown of Rho GDIα. [Breast Cancer Res Treat] Abstract

Effect of Docosahexanoic Acid on Cell Cycle Pathways in Breast Cell Lines with Different Transformation Degree
Following treatments with docosahexanoic acid scientists examined cell viability, death, cell cycle and some molecular effects in breast cell lines with different transformation, phenotypic and biochemical characteristics. [J Cell Physiol] Abstract

Phenethyl Isothiocyanate, by Virtue of Its Antioxidant Activity, Inhibits Invasiveness and Metastatic Potential of Breast Cancer Cells: HIF-1α as a Putative Target
Researchers explored the effect of phenethyl isothiocyanate, a natural isothiocyanate, found in cruciferous vegetables on the expression of hypoxia-inducible factor 1α (HIF-1α) and HSP90 in breast adenocarcinoma cell lines under both normoxia and hypoxia. [Free Radic Res] Abstract

Involvement of Gelsolin in TGF-Beta 1 Induced Epithelial to Mesenchymal Transition in Breast Cancer Cells
Gelsolin (GSN) overexpression in both MDA-MB231 and MCF-7 cells demonstrated that increased GSN expression resulted in alterations of cell proliferation and cell cycle progression, modification of the actin filament assembly associated with altering cell surface elasticity and cell detachment in these breast cancer cells. [J Biomed Sci] Abstract

The Human Chemokine Receptor CCRL2 Suppresses Chemotaxis and Invasion by Blocking CCL2-Induced Phosphorylation of p38 MAPK in Human Breast Cancer Cells
Scientists investigated whether CCRL2 is involved in the chemotaxis, invasion, and proliferation of human breast cancer cells. Expression of CCRL2 was determined in six breast cancer cell lines by real-time RT-PCR and Western blot. [Med Oncol] Abstract

Leptin and Adiponectin Modulate the Self-Renewal of Normal Human Breast Epithelial Stem Cells
Researchers investigated the effects of leptin and adiponectin on normal breast epithelial stem cells. Levels of leptin in human adipose explant-derived conditioned media positively correlated with the size of the normal breast stem cell pool. [Cancer Prev Res] Abstract

    CLINICAL RESEARCH

Etirinotecan Pegol (NKTR-102) versus Treatment of Physician’s Choice in Women with Advanced Breast Cancer Previously Treated with an Anthracycline, a Taxane, and Capecitabine (BEACON): A Randomized, Open-Label, Multicenter, Phase III Trial
The authors assessed whether etirinotecan pegol is superior to currently available treatments for patients with previously treated, locally recurrent or metastatic breast cancer. [Lancet Oncol] Abstract

A Phase II Study of Capecitabine plus Cisplatin in Metastatic Triple-Negative Breast Cancer Patients Pretreated with Anthracyclines and Taxanes
Investigators evaluated the efficacy and safety of capecitabine plus cisplatin regimen in metastatic triple-negative breast cancer patients pretreated with anthracyclines and taxanes. [Cancer Biol Ther] Abstract

    REVIEWS
Biological Markers of Invasive Breast Cancer
In recent years, gene expression profiling has made the molecular classification of breast cancer possible. Classification of breast cancer by immunohistochemical expression of estrogen receptor, progesterone receptor, human epidermal growth factor receptor 2 and Ki-67 is standard practice for clinical decision-making. [Jpn J Clin Oncol] Abstract

    INDUSTRY NEWS
OncoQR’s Breast Cancer Vaccine Successfully Completes Pre-Clinical Proof-of-Concept Study
OncoQR ML announced that its breast cancer vaccine candidate OQR200 has successfully completed a proof-of-concept study in non-human primates. The vaccine elicited the expression of very high levels of polyclonal antibodies targeting multiple epitopes on Her2/neu. [OncoQR ML (B3C Group GmbH)] Press Release

Stem Cell Researcher Min Yu Receives the NIH’s New Innovator Award
As a winner of the NIH Director’s New Innovator Award, USC Stem Cell principal investigator Min Yu will strive to develop individualized medicine targeting rare and deadly breast cancer stem cells. The five-year, $2.475 million award is part of the High-Risk, High-Reward Research program supported by the NIH Common Fund. [Keck School of Medicine of USC] Press Release

Do Stem Cells Hold the Key to Breast Cancer Spread?
A $680,000 investment in breast cancer research is helping Dr. Ewa Michalak to find the stem cells that hold the key to how breast cancer spreads, and could lead to better treatments for the disease. [The Walter and Eliza Hall Institute of Medical Research] Press Release
