HER2 Missense Mutations Have Distinct Effects on Oncogenic Signaling and Migration
Scientists functionally characterized human epidermal growth factor receptor 2 (HER2) kinase and extracellular domain mutations through gene editing of the endogenous loci in HER2 nonamplified human breast epithelial cells. [Proc Natl Acad Sci USA] Abstract

PUBLICATIONS (Ranked by impact factor of the journal)
    LABORATORY RESEARCH

HIF-1 Regulates CD47 Expression in Breast Cancer Cells to Promote Evasion of Phagocytosis and Maintenance of Cancer Stem Cells
Researchers report that hypoxia-inducible factor 1 (HIF-1) directly activates transcription of the CD47 gene in hypoxic breast cancer cells. [Proc Natl Acad Sci USA] Abstract

Aberrantly Upregulated TRAP1 Is Required for Tumorigenesis of Breast Cancer
Investigators showed that tumor necrosis factor receptor-associated protein 1 (TRAP1) is aberrantly upregulated in breast tumors compared to control tissues. [Oncotarget] Full Article

Trastuzumab Resistance Induces EMT to Transform HER2+ PTEN− to a Triple Negative Breast Cancer that Requires Unique Treatment Options
Researchers demonstrated that continued use of trastuzumab in HER2+ cells with loss of PTEN induces the epithelial-mesenchymal transition (EMT) and transform HER2+ to a triple negative breast cancer. [Sci Rep] Full Article

Bub1 Is Required for Maintaining Cancer Stem Cells in Breast Cancer Cell Lines
Scientists identified that depleting Bub1 (Budding uninhibited by benzimidazoles 1) using shRNAs reduces cancer stem cell potential of the MDA-MB-231 breast cancer cell line, resulting in inhibited formation of xenografts in immunocompromised mice. [Sci Rep] Full Article

Tumor Cell-Derived Periostin Regulates Cytokines that Maintain Breast Cancer Stem Cells
Researchers demonstrated that an intact periostin-integrin beta3 signaling axis is required for the maintenance of breast cancer stem cells. [Mol Cancer Res] Abstract

The Breast Cancer Susceptibility Gene Product (γ-Synuclein) Alters Cell Behavior through Its Interaction with Phospholipase Cβ
Using cells that mimic stage 4 breast cancer, the authors showed that down-regulation of γ-synuclein reduces the protein level of PLCβ but increases the transcript level over 40 fold. [Cell Signal] Abstract

miR-33a Suppresses Breast Cancer Cell Proliferation and Metastasis by Targeting ADAM9 and ROS1
Researchers found that downregulated miR-33a in breast cancer tissues correlates with lymph node metastasis. miR-33a expression was significantly lower in the highly metastatic breast cancer cell lines than the noncancerous breast epithelial cells and non-metastatic breast cancer cells. [Protein Cell] Full Article

Methyl Sulfone Blocked Multiple Hypoxia- and Non-Hypoxia-Induced Metastatic Targets in Breast Cancer Cells and Melanoma Cells
Investigators showed that the molecule, methyl sulfone, normalized metastatic metabolism of hypoxic breast cancer and melanoma cells by altering several metabolic functions of the cells. [PLoS One] Full Article

    CLINICAL RESEARCH

Molecular Heterogeneity and Response to Neoadjuvant Human Epidermal Growth Factor Receptor 2 Targeting in CALGB 40601, a Randomized Phase III Trial of Paclitaxel plus Trastuzumab with or without Lapatinib
CALGB 40601 examined the impact of dual HER2 blockade consisting of trastuzumab and lapatinib added to paclitaxel, considering tumor and microenvironment molecular features. [J Clin Oncol] Full Article
