TOP STORYDiscovery of a Periosteal Stem Cell Mediating Intramembranous Bone Formation Investigators identified a periosteal stem cell that was present in the long bones and calvarium of mice, displayed clonal multipotency and self-renewal, and sat at the apex of a differentiation hierarchy. PUBLICATIONS(Ranked by impact factor of the journal)IN VITROMesenchymal stem cells are considered to be an attractive cell type for therapeutic exosome production. To optimize exosome production, researchers cultivated umbilical cord-derived mesenchymal stem cells in scalable microcarrier-based three-dimensional cultures. [Mol Ther] Abstract Scientists reported that basic fibroblast growth factor (FGF2)-Ras homolog gene family member A (RhoA)/Rho-associated protein kinase (ROCK) signaling functioned as a molecular switch to regulate the lineage fate of bone MSCs and that prevascularization promoted the cell fate switch, which contributed to increased bone regeneration with the use of prevascularized tissue-engineered bone grafts (TEBGs) compared with control TEBGs. [Cell Death Dis] Full Article To better identify seed cells for bone tissue engineering, investigators compared MSCs from seven different tissues including four from dental origins, two from somatic origins and one from birth-associated perinatal tissue umbilical cord. MSCs from dental and perinatal tissues proliferated more rapidly than those from somatic origins. [Stem Cells Dev] Abstract Co-expression of Akt1 and Wnt11 markedly promoted the expression of cardiac markers including NK2 transcription factor related 5, GATA-binding protein 4, α-myosin MHC and brain natriuretic protein. Co-expression of Akt1 and Wnt11 increased cell survival and reduced cell apoptosis of MSCs under hypoxia/reoxygenation treatment; however, these effects were blocked by Wnt11 neutralizing antibodies or Akt1 inhibitor. [Biomed Pharmacother] Abstract Low and medium accelerations enhanced osteogenesis in mesenchymal stem cells while high accelerations inhibited it. [J Tissue Eng] Full Article IN VIVOIdentification of the Human Skeletal Stem Cell Researchers reported the isolation of a self-renewing and multipotent human skeletal stem cell that generated progenitors of bone, cartilage, and stroma, but not fat. [Cell] Full Article | Press Release | Graphical Abstract | Video Investigators reported that regardless of risk-group, bone-marrow MSCss from acute myeloid leukemia patients supported, similar to those derived from healthy bone-marrow the survival, proliferation, differentiation and clonogenecity of CD34+ cells in vitro, and the in vivo immune deficient mice repopulating assays. [Haematologica] Abstract | Full Article Notch Ligand Jagged1 Promotes Mesenchymal Stromal Cell-Based Cartilage Repair In vivo cell tracing showed that more Jagged1 (JAG1)-treated placenta-derived MSCs (PMSCs) remained in knee joint tissues and that JAG1-treated PMSCs exhibited greater PMSC chondrogenic differentiation than lgG-treated control PMSCs at four weeks after injection. POLICY NEWSTrump Administration Launches Review of Scientific Research Involving Fetal Tissue The Trump administration has launched a comprehensive review of all research that involves fetal tissue, reopening an issue that has galvanized anti-abortion activists but worried scientists who fear their work could be under threat. [STAT News] Editorial Portrayals of Female STEM Characters in TV and Film Haven’t Improved in 10 Years The Lyda Hill Foundation, in partnership with Geena Davis’s Institute on Gender in Media at Mount Saint Mary’s University, released an extensive research report on the portrayals of female characters in science, technology, engineering and math in television and film. “Portray Her: Representations of Women STEM Characters in Media” shows that entertainment media has a long way to go to improve stereotypes about pursuing STEM careers.
