TOP STORYResearchers demonstrated that N-cadherin overexpression enhanced MSC protective effects against ischemic heart failure via β-catenin mediated MMP-10/MMP-13/HGF expression and production, promoting adipose tissue-derived MSC (ADSC)/cardiomyocyte adhesion and ADSC retention. PUBLICATIONS(Ranked by impact factor of the journal)IN VITROStepwise osteogenesis-mimicking extracellular matrix (ECM) deposited poly (lactic-coglycolic acid) (PLGA)-collagen hybrid meshes showed different ECM compositions associated with the stage of osteogenesis, their effects on the osteogenic differentiation of human MSCs (hMSCs) differed. Early stage-ECM scaffold increased and late stage-ECM scaffold moderately promoted the osteogenic differentiation of hMSCs. [Biofabrication] Abstract Scientists found miR-765 overexpression to inhibit osteogenesis-associated gene upregulation during osteogenic differentiation of human MSCs, whereas knockdown of this miRNA was associated with increased expression of these genes. [Stem Cell Res Ther] Full Article Investigators report KDM4D histone demethylase as a key regulator of adipogenesis in C3H10T1/2 MSCs. The depletion of KDM4D resulted in impaired differentiation, which could be rescued by exogenous KDM4D, PPARγ, and C/EBPα, but not by C/EBPβ. [Sci Rep] Full Article Silk-Reinforced Collagen Hydrogels with Raised Multiscale Stiffness for Mesenchymal Cells 3D Culture The authors present a new, simple and effective method to develop a collagen-silk hybrid hydrogel with high collagen concentration and with increased stiffness approaching that of natural tissues, which could be of high interest for the development of cardiac patches for myocardial regeneration and for preconditioning of MSCs to improve their therapeutic potential. [Tissue Eng Part A] Abstract IN VIVOScientists tested the efficacy of the photoactivation of MSCs loaded with nanoparticles in vitro and in a murine in vivo ectopic osteosarcoma model. [J Exp Clin Cancer Res] Full Article MSCs infused immediately and 24 hours later both markedly ameliorated myocardial ischemia‐reperfusion injury, and immediate infusion had more significant outcome. These improvements were associated with neutrophil infiltration, measured by fluorescence-activated cell sorting analysis and immunofluorescence staining. [J Am Heart Assoc] Full Article In an in vivo experiment, single-cell adipose-derived stem cells (ADSCs) and 3D-cultured ADSC spheroids were injected intra-articularly into the knees of rheumatoid arthritis model mice and histologically assessed. Marked improvement of synovial inflammation and articular cartilage regeneration was found in ADSC-treated mice. [Sci Rep] Full Article A new thermosensitive composite hydrogel based on chitosan, hydroxyethyl cellulose, collagen, and β-phosphoglycerate was developed to encapsulate murine bone marrow-derived MSCs for improving the therapeutic efficacy of spinal cord injury mice. [Biomed Mater] Abstract Researchers showed that β3 adrenergic agonists (β3AR) in combination with a CXCR4 antagonist, AMD3100/Plerixafor, could mobilize MSCs into the blood in mice and rats. Mechanistically they showed that reversal of the CXCL12 gradient across the bone marrow endothelium and local generation of endocannabinoids may both play a role in this process. INDUSTRY NEWSThe Center for International Blood and Marrow Transplant Research® (CIBMTR®) announced the selection of IQVIA as it seeks to expand the value real world data can bring to patients, through systematic investigation to generate real world evidence. The CIBMTR is a long-standing research collaboration between the National Marrow Donor Program®/Be The Match® and the Medical College of Wisconsin, dedicated to improving survival, treatment and quality of life for transplant and cellular therapy patients. [CIBMTR®] Press Release Mesoblast Limited announced that the investigator-initiated expanded access protocol using its cryopreserved allogeneic cell therapy product candidate remestemcel-L for steroid-refractory chronic graft versus host disease has resulted in clinically meaningful outcomes in all three treated patients, two children and one adult, within 28 days after two infusions. POLICY NEWSMeTooSTEM Organization Reeling after More Resignations The prominent nonprofit group MeTooSTEM appears to be imploding amid resignations and accusations of bullying by founder BethAnn McLaughlin. Critics, many of whom are antiharassment advocates, say she has sidelined people of color and bullied volunteers, activists, and fellow leaders both at the nonprofit and outside of it. [ScinceInsider] Editorial White House Formally Invites Public Comment on Open-Access Policies The White House issued a notice Wednesday seeking comment on its effort to enhance public access to federally funded research. It’s an old idea creating new controversy. White House Office of Science and Technology Policy Director Kelvin Droegemeier is pushing back against publishers that in December said the administration was quietly pursuing an executive order to require immediate free distribution of taxpayer funded research. [ScienceInsider] Editorial Has the UK Science Minister Been Demoted? A government reshuffle in the United Kingdom has born another new science minister – a position that has seen a revolving door of appointees, firings and resignations in recent years. The latest appointment also comes with an apparent political demotion for the role, which expert observers say is in conflict with a government that appears to have put research and innovation at the heart of its agenda.
