TOP STORY Rb Regulates Fate Choice and Lineage Commitment In Vivo Because osteoblasts and adipocytes can both arise from mesenchymal stem cells, observations suggest that pRb might play a role in the choice between these two fates. However, so far, there is no evidence for this in vivo . Here researchers used mouse models to address this hypothesis in mesenchymal tissue development and tumorigenesis. [Nature]

SCIENCE NEWS Newts’ Ability to Regenerate Tissue Replicated in Mouse Cells by Scientists Scientists have taken a big step toward being able to confer newts’ regenerative capacity on mammalian muscle cells; they accomplished this feat in experiments with laboratory mice in which they blocked the expression of just two tumor-suppressing proteins. [Press release from Stanford University discussing online prepublication in Cell Stem Cell] New Insights into How Stem Cells Determine What Tissue to Become Within 24 hours of culturing adult human stem cells on a new type of matrix, researchers were able to make predictions about how the cells would differentiate, or what type of tissue they would become. [Press release from the University of Michigan discussing online prepublication in Nature Methods]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) Transient Inactivation of Rb and ARF Yields Regenerative Cells from Postmitotic Mammalian Muscle The study’s results show that differentiation of mammalian cells is reversed by inactivation of Arf and Rb and supported the hypothesis that Arf evolved at the expense of regeneration. [Cell Stem Cell] Mechanical Regulation of Cell Function with Geometrically Modulated Elastomeric Substrates Scientists reported the establishment of a library of micromolded elastomeric micropost arrays to modulate substrate rigidity independently of effects on adhesive and other material surface properties. [Nat Methods] Bone Marrow Mesenchymal Stem Cells Stimulate Cardiac Stem Cell Proliferation and Differentiation Here, researchers tested the hypothesis that bone marrow derived mesenchymal stem cells stimulate the proliferation and differentiation of endogenous cardiac stem cells as part of their regenerative repertoire. [Circ Res] The Role of Biomaterials in the Direction of Mesenchymal Stem Cell Properties and Extracellular Matrix Remodeling in Dermal Tissue Engineering This study compared the epithelial-mesenchymal interaction and extracellular matrix remodeling of cell-based and collagen-based dermal equivalents using fibroblasts, human mesenchymal stem cells from bone marrow or Wharton’s Jelly of the umbilical cord, respectively, in co-culture with the keratinocyte cell line HaCaT. [Biomaterials] The BRG1 ATPase of Chromatin Remodeling Complexes is Involved in Modulation of Mesenchymal Stem Cell Senescence Through RB-P53 Pathways Investigators focused their attention on brahma-related gene 1, the ATPase subunit of the SWItch/Sucrose NonFermentable chromatin remodeling complex, and analyzed its role in mesenchymal stem cell biology. [Oncogene] Proteome Analysis of Rat Bone Marrow Mesenchymal Stem Cell Differentiation This study was designed to investigate the protein expression profiles of rat bone marrow mesenchymal stem cells during differentiation into adipogenic (by dexamethasone, isobutylmethylxanthine, insulin and indomethacin), cardiomyogenic (by 5-azacytidine), chondrogenic (by ascorbic acid, insulin-transferrin-selenous acid, and transforming growth factor-beta 1) and osteogenic (by dexamethasone, beta-glycerophosphate, and ascorbic acid) lineages by well-known differentiation inducers. [J Proteome Res] Tissue-Resident Mesenchymal Stem Cells Attract Peripheral Blood Neutrophils and Enhance Their Inflammatory Activity in Response to Microbial Challenge Scientists hypothesized that tissue-specific mesenchymal stem cells (MSCs) could play an instructional role during early phases of bacterial challenge. To investigate this hypothesis further, scientists set up a coculture system of glandular MSCs and peripheral blood neutrophils so that they could analyze the cellular interactions of these cells in response to LPS challenge. [J Leukoc Biol] Oxygen Tension Differentially Regulates the Functional Properties of Cartilaginous Tissues Engineered from Infrapatellar Fat Pad Derived MSCs and Articular Chondrocytes The objective of this study was to investigate how chondrocytes and infrapatellar fad pad derived mesenchymal stem cells will respond to a low oxygen (5%) environment in 3D agarose culture. The hypothesis was that culture in a low oxygen environment (5%) will enhance the functional properties of cartilaginous tissues engineered using both cell sources. [Osteoarthristis Cartilage]
