    TOP STORY
The Influence of Delayed Compressive Stress on TGF-ß1-Induced Chondrogenic Differentiation of Rat BMSCs through Smad-Dependent and Smad-Independent Pathways
Scientists have previously shown that Col2α1 mRNA was increased by delayed dynamic compressive stress initiated at the eighth day of chondrogenic culture. Here, they further studied the possibility of using delayed mechanical stress to relay chondrogenesis initiated by exogenous transforming growth factor-ß1 (TGF-ß1). [Biomaterials] Abstract

Inducing Ossification in an Engineered Three-Dimensional Scaffold-Free Living Cartilage Template
Scientists employed an in vitro macro-sized three-dimensional cell-based platform for investigation and application of osteogenesis. To evaluate the efficiency of living hyaline cartilaginous graft (LhCG) as a viable platform for bone formation, osteoblast and human mesenchymal stem cells were seeded respectively into LhCG constructs, establishing a co-culture system consisting of osteo-progenitors and chondrocytes. [Biomaterials] Abstract

Human Amniotic Fluid Stem Cell Injection Therapy for Urethral Sphincter Regeneration in an Animal Model
Researchers found that human amniotic fluid stem cells (hAFSCs) displayed mesenchymal stem cell characteristics and could differentiate into cells of myogenic lineage. Periurethral injection of hAFSCs into an stress urinary incontinence animal model restored the urethral sphincter to apparently normal histology and function, in absence of immunogenicity and tumorigenicity. Activated NF-kB in Bone Marrow Mesenchymal Stem Cells from SLE Patients Inhibits Osteogenic Differentiation through Down-Regulating Smad Signaling
Researchers investigated the bone morphogenetic protein-2 (BMP-2) induced osteoblastic capacity of bone marrow-derived mesenchymal stem cells from systemic lupus erythematosus (SLE) patients and the tumor necrosis factor signaling system in determining BMP-2-induced the regulatory pathways. [Stem Cells Dev] Abstract

Mesenchymal Stem Cells Attenuate Peritoneal Injury through Secretion of TNFα-Stimulating Gene-6
Scientists investigated the effects of mesenchymal stem cells on injured mesothelial cells and peritoneal fibrosis. [PLoS One] Full Article

Human Mesenchymal Stem Cell Expression Program upon Extended Ex Vivo Cultivation, as Revealed by 2-DE-Based Quantitative Proteomics
To get clues into the molecular mechanisms underlying cellular senescence resulting from extended ex vivo cultivation of bone marrow mesenchymal stem cells, researchers explored a two-dimensional gel electrophoresis (2-DE) based quantitative proteomics to compare the expression programs of Passage 3 cells (P3) and P7 cells. [PLoS One] Full Article

Neural Differentiation Ability of Mesenchymal Stromal Cells from Bone Marrow and Adipose Tissue: A Comparative Study
The proliferation capacity, secretion ability of neurotrophins and neural differentiation ability in rat mesenchymal stromal cells isolated from bone marrow and adipose tissue were compared from the same animal. [Cytotherapy] Abstract

Analysis of In Vitro Secretion Profiles from Adipose-Derived Cell Populations
Investigators found that the use of mixed adipose cell populations does not appear to induce cellular stress and results in enhanced secretion profiles. [J Transl Med] Abstract | Full Article

Adipose Stem Cells Can Secrete Angiogenic Factors That Inhibit Hyaline Cartilage Regeneration
Scientists aimed to determine the effects adipose stem cell-secreted factors have in repairing chondral defects. [Stem Cell Res Ther] Abstract | Full Article | Press Release

Macrophages Modulate the Viability and Growth of Human Mesenchymal Stem Cells
Researchers investigated how cytokines and polarized macrophages associated with pro-inflammatory and anti-inflammatory responses affect the survival of mesenchymal stem cells. [J Cell Biochem] Abstract

Intra-Articular Delivery of Purified Mesenchymal Stem Cells from C57BL/6 or MRL/MpJ Superhealer Mice Prevents Post-Traumatic Arthritis
Researchers determined whether the delivery of mesenchymal stem cells (MSCs) would prevent post-traumatic arthritis by altering the balance of inflammation and regeneration after fracture of the mouse knee. They also examined the whether MSCs from the MRL/MpJ “superhealer” mouse strain would show increased multilineage and therapeutic potentials as compared to those from C57BL/6 mice. [Cell Transplant] Abstract

Isolation, Characterization, and Differentiation of Stem Cells for Cartilage Regeneration
The authors identify current approaches for isolating and chondrogenically differentiating mesenchymal stem cells from bone marrow, fat, synovium, muscle, and peripheral blood, as well as cells from extra-embryonic tissues, embryonic stem cells, and induced pluripotent stem cells. They also assess chondrogenic induction with growth factors, identifying standard cocktails used for each stem cell type. [Ann Biomed Eng] Abstract

The Maryland Stem Cell Research Commission: Public Statement
The Maryland Stem Cell Research Commission is pleased with the opinion issued by the U.S. Court of Appeals for the District of Columbia Circuit in Sherley, et al vs. Sebelius, et al regarding the federal funding of human embryonic stem cell research. The position taken by the Court allows this promising stem cell research to progress in the U.S., paving the way for groundbreaking discoveries that could ameliorate a multitude of debilitating diseases and disorders.
