    TOP STORY
Ostrow Study Identifies Mechanism that Makes Ordinary Stem Cells Create Tumors
A new study illustrates how changes in cell signaling can cause ordinary stem cells in the jaw to start forming benign but potentially harmful tumors. [Press release from University of Southern California discussing online prepublication in Cell Stem Cell]

Mesenchymal Stem Cells Cancel Azoxymethane-Induced Tumor Initiation
Scientists determined whether exogenous mesenchymal stem cells possess intrinsic antineoplastic or proneoplastic properties in azoxymethane (AOM)-induced carcinogenesis. Three in vivo models were studied: an AOM/dextran sulfate sodium colitis-associated carcinoma model, an aberrant crypt foci model, and a model to assess the acute apoptotic response of a genotoxic carcinogen. [Stem Cells ] Abstract

Insulin-Like Growth Factor Binding Proteins 4 and 7 Released by Senescent Cells Promote Premature Senescence in Mesenchymal Stem Cells
Researchers showed that conditioned media of senescent mesenchymal stem cells contain a set of secreted factors that are able to induce a full senescence response in young cells. [Cell Death Dis] Full Article

CD146 Expression on Mesenchymal Stem Cells Is Associated with Their Vascular Smooth Muscle Commitment
CD146-/Low and CD146High bone marrow mesenchymal stem cells did not differ in colony-forming unit-fibroblast number, osteogenic, adipogenic and chondrogenic differentiation or in vitro hematopoietic-supportive activity. [J Cell Mol Med] Full Article

The Paracrine Effects of Mesenchymal Stem Cells Stimulate the Regeneration Capacity of Endogenous Stem Cells in the Repair of a Bladder Outlet Obstruction-Induced Overactive Bladder
Scientists report the superior therapeutic efficacy of using mesenchymal stem-cells for the treatment of OAB and a novel therapeutic mechanism that activates endogenous Oct4+ primitive stem-cells. [Stem Cells Dev] Abstract

CC-Chemokine Receptor 5 on Pulmonary Mesenchymal Cells Promotes Experimental Metastasis via the Induction of Erythroid Differentiation Regulator 1
Though commonly associated with bone marrow stroma, Erythroid Differentiation Regulator 1 was differentially expressed in wild type pulmonary mesenchymal cells and murine embryonic fibroblasts. [Mol Cancer Res] Abstract

A Fat Option for the Pig: Hepatocytic Differentiated Mesenchymal Stem Cells for Translational Research
Mesenchymal stem cells (MSC) from porcine adipose tissue and from bone marrow express the typical mesenchymal markers CD44, CD29, CD90 and CD105 but not hematopoietic markers. MSC from both sources displayed differentiation into the osteogenic as well as adipogenic lineage. [ Exp Cell Res] Abstract

Inhibition of Non-Muscle Myosin II Leads to G0/G1 Arrest of Wharton’s Jelly-Derived Mesenchymal Stromal Cells
When cultured in the presence of blebbistatin, an inhibitor of non-muscle myosin II adenosine triphosphatase activity, Wharton’s jelly-derived mesenchymal stromal cells exhibited dose-dependent reduction in proliferative potential along with increase in cell size and induction of early senescence. [Cytotherapy] Abstract

Gastritis Promotes an Activated Bone Marrow-Derived Mesenchymal Stem Cell with a Phenotype Reminiscent of a Cancer-Promoting Cell
In culture, bone marrow-derived mesenchymal stem cells (BM-MSCs) are prone to mutation with continued passage but it is unknown whether a similar process occurs in vivo in response to gastritis. Scientists identified the role of chronic gastritis in the transformation of BM-MSCs leading to an activated cancer-promoting phenotype. [ Dig Dis Sci] Abstract

Differentiation of Human Mesenchymal Stem Cells on Plasma-Treated Polyetheretherketone
To evaluate the effect of the plasma-induced polyetheretherketone (PEEK) modifications on stem cell adhesion and differentiation, adipose tissue-derived mesenchymal stem cells (adMSC) were seeded on PEEK specimens. Scientists demonstrated an increased adhesion, proliferation, and osteogenic differentiation of adMSC in contact to plasma-treated PEEK. [J Mater Sci Mater Med] Abstract

Aorta-Derived Mesoangioblasts Can Be Differentiated into Functional Uterine Epithelium, but Not Prostatic Epithelium or Epidermis, by Instructive Mesenchymes
Researchers examined whether aorta-derived mesoangioblasts (ADM) can differentiate into prostatic, uterine, and skin epithelium by recombining ADM with fetal or neonatal mesenchyme from these organs and grafting them under the renal capsule of syngeneic hosts. [Cells Tissues Organs] Abstract

Deciphering Mechanical Regulation of Chondrogenesis in Fibrin-Polyurethane Composite Scaffolds Enriched with Human Mesenchymal Stem Cells; A Dual Computational and Experimental Approach
Researchers used the finite element method, mechanical testing and dynamic in-vitro cell culture experiments on human bone marrow derived mesenchymal stem cells-enriched fibrin-polyurethane composite scaffolds in order to quantitatively decipher the mechanoregulation of chondrogenesis within fibrin-polyurethane composite scaffold constructs. [Tissue Eng Part A] Abstract

Significance of Calcium Phosphate Coatings for the Enhancement of New Bone Osteogenesis – A Review
It has been demonstrated that calcium phosphate (CaP) coatings have a significant effect on the bone regeneration process. In vitro experiments using different cells have revealed that CaP coatings enhance cellular adhesion, proliferation, and differentiation to promote bone regeneration. [Acta Biomater] Abstract

Tumor Microenvironment: Bone Marrow-Mesenchymal Stem Cells as Key Players
The authors discuss the role of bone marrow-derived mesenchymal stem cells (BM-MSC) in many steps in tumor progression, focusing on the factors that attract BM-MSC to tumors, BM-MSC differentiation ability, the role of BM-MSC in tumor support or inhibition, the immunomodulation promoted by BM-MSC and metastatic niche formation by these cells. [Biochim Biophys Acta] Abstract

Mesoblast’s Mesenchymal Precursor Cells Induce Functional Recovery in Ischemic Stroke Model
Mesoblast Limited announced positive preclinical trial results which demonstrated that its proprietary Mesenchymal Precursor Cells (MPCs) could improve functional recovery following a stroke. The trial results showed that a single intravenous injection of human MPCs significantly enhanced sensorimotor recovery when administered up to seven days after an ischemic stroke in rats.
