    TOP STORY
Research Team Pursues Techniques to Improve Elusive Stem Cell Therapy
Researchers found that transplanting mesenchymal stem cells along with blood vessel-forming cells naturally found in circulation improves stem cell survival in mice after engraftment. [Press release from Harvard Stem Cell Institute discussing online prepublication in the Proceedings of the National Academy of Sciences]

Amino-Polyvinyl Alcohol Coated Superparamagnetic Iron Oxide Nanoparticles Are Suitable for Monitoring of Human Mesenchymal Stromal Cells In Vivo
After identifying non-interfering cell-assays and determining the delivered and cellular dose, in addition to the administered dose, amino-polyvinyl alcohol coated super-paramagnetic iron-oxide nanoparticles (A-PVA-SPIONs) were found to be non-toxic to mesenchymal stromal cells (MSCs) and non-destructive towards their multi-lineage differentiation potential. Surprisingly, MSC migration was increased. In magnetic resonance imaging, A-PVA-SPION-labeled MSCs were successfully visualized in vitro and in vivo. [Small] Abstract

Leptin Signaling Is Required for Augmented Therapeutic Properties of Mesenchymal Stem Cells Conferred by Hypoxia Preconditioning
The mechanism underlying hypoxia-induced augmentation of the protective effect of mesenchymal stem cells on myocardial infarction is poorly understood. Scientists show that hypoxia-enhanced survival, mobility, and protection of co-cultured cardiomyocytes were paralleled by increased expression of leptin and cell surface receptor CXCR4. [Stem Cells] Abstract

Biological Markers of Mesenchymal Stromal Cells as Predictors of Response to Autologous Stem Cell Transplantation in Patients with Amyotrophic Lateral Sclerosis; An Investigator-Initiated Trial and In Vivo Study
This open-label, single-arm, investigator-initiated clinical study was designed to identify markers of mesenchymal stromal cells (MSCs) that could be used as potential predictors of response to autologous MSC therapy in patients with amyotrophic lateral sclerosis. [Stem Cells] Abstract

Chronic Restraint Stress Decreases the Repair Potential from Mesenchymal Stem Cells on Liver Injury by Inhibiting TGF-β1 Generation
Researchers illustrated that mesenchymal stem cells (MSCs) could repair liver injury by differentiating into myofibroblasts (MFs) which contribute to fibrosis, whereas stress repressed differentiation of MSCs into MFs displayed by reducing α-smooth muscle actin expression. In Vitro Osteogenic Differentiation of Adipose-Derived Mesenchymal Stem Cell Spheroids Impairs Their In Vivo Vascularization Capacity Inside Implanted Porous Polyurethane Scaffolds
Undifferentiated adipose-derived mesenchymal stem cell spheroids are attractive vascularization units for tissue engineering. Scientists analyzed how differentiation affects their in vivo vascularization capacity. [Acta Biomater] Abstract

Mesenchymal Stem Cells Induce the Ramification of Microglia via the Small RhoGTPases Cdc42 and Rac1
Researchers investigated the effect of adipose-tissue-derived mesenchymal stem cells (ASCs), which exert important anti-inflammatory actions, in microglia morphology. Microglia exposed to ASCs or their secreted factors underwent a cell shape change into a ramifying morphology in basal and inflammatory conditions, similar to that observed in microglia found in healthy brain. [Glia] Abstract

Adipose-Derived Stem Cells Retain Their Regenerative Potential after Methotrexate Treatment
Scientists investigated the effects of chemotherapeutics on adipose-derived stem cell function to determine whether this cell source could be a candidate for repairing, or even preventing, chemotherapy-induced tissue damage. [Exp Cell Res] Abstract

Neurotrophic and Neuroprotective Potential of Human Limbus-Derived Mesenchymal Stromal Cells
Scientists examined neurotrophic and neuroprotective effects of limbus stroma-derived mesenchymal stromal cells on cortical neurons in vitro and in vivo. [Cytotherapy] Abstract

A Comparison of the In Vitro Mineralization and Dentinogenic Potential of Mesenchymal Stem Cells Derived from Adipose Tissue, Bone Marrow and Dental Pulp
Scanning electron microscopy and alizarin red staining showed that the pattern of mineralization in dental pulp stem cell (DPSC) cultures differed from adipose tissue stem cells and bone marrow stem cells, with DPSC cultures lacking defined mineralized nodules and instead forming a diffuse layer of low-density mineral. [J Bone Miner Metab] Abstract

Growth on Poly(l-Lactic Acid) Porous Scaffold Preserves CD73 and CD90 Immunophenotype Markers of Rat Bone Marrow Mesenchymal Stromal Cells
Researchers studied the effect of porous, flat and smooth poly(l-lactic acid) scaffolds on the immunophenotype of in vitro grown, rat mesenchymal stromal cells in the absence of any coating, co-polymeric enrichment, and differentiation stimuli. [J Mater Sci Mater Med] Abstract

Whole-Genome Sequencing Identifies Genetic Variances in Culture-Expanded Human Mesenchymal Stem Cells
Researchers present a whole-genome investigation on the genetic dynamics of cultured mesenchymal stem cells under ex vivo establishment and serial expansion. [Stem Cell Rep] Full Article | Graphical Abstract

Umbilical Cord Blood Mesenchymal Stem Cells Co-Modified by TERT and BDNF: A Novel Neuroprotective Therapy for Neonatal Hypoxic-Ischemic Brain Damage
Compared with other types of stem cells, umbilical cord blood mesenchymal stem cells (UCB-MSCs) have several unique characteristics, such as a higher rate of cell proliferation and clonality. However, the limited life span of UCB-MSCs hinders their clinical application. Therefore, efforts are urgently needed to circumvent this disadvantage. [Int J Dev Neurosci] Abstract
