    Scientists have identified a new therapeutic approach that, while still preliminary, could promote the development of new bone-forming cells in patients suffering from bone loss.     Oncogenic Transformation Can Orchestrate Immune Evasion and Inflammation in Human Mesenchymal Stem Cells Independently of Extrinsic Immune-Selective Pressure
    Neoplastic transformation increased the natural immune evasive properties of human mesenchymal stem cells (hMSC), both by reducing their immunogenicity and by increasing their capacity to inhibit mitogen driven T-cell proliferation. Researchers also found that IFN-γ signaling was globally affected in transformed hMSC. [Cancer Res] Abstract

    Simple Coating with Fibronectin Fragment Enhances Stainless Steel Screw Osseointegration in Healthy and Osteoporotic Rats
    The authors present a simple coating strategy based on passive adsorption of FN7-10, a recombinant fragment of human fibronectin encompassing the major cell adhesive, integrin-binding site, onto 316-grade stainless steel (SS). FN7-10 coating on SS surfaces promoted α5β1 integrin-dependent adhesion and osteogenic differentiation of human mesenchymal stem cells. [Biomaterials] Abstract

    SWI/SNF-Mediated Lineage Determination in Mesenchymal Stem Cells Confers Resistance to Osteoporosis
    BRM is not required for development, which has hindered understanding of its contributions, but knockdown genetics, designed to explore the hypothesis that BRM-SWI/SNF has different regulatory roles in different mesenchymal stem cell lineages, showed that depleting BRM from mesenchymal stem cells has a dramatic effect on the balance of lineage selection between osteoblasts and adipocytes.     A Method for Performing Islet Transplantation Using Tissue-Engineered Sheets of Islets and Mesenchymal Stem Cells
    Investigators describe a tissue engineering approach using islets with mesenchymal stem cell sheets to improve the therapeutic effect of islet transplantation. [Tissue Eng Part C Methods] Abstract

    Gata4, Tbx5 and Baf60c Induce Differentiation of Adipose Tissue-Derived Mesenchymal Stem Cells into Beating Cardiomyocytes
    Scientists showed that the combination of Gata4, Tbx5 and Baf60c is sufficient for inducing adipose tissue-derived mesenchymal stem cells to form cardiomyocytes. It also appears that, while Gata4 and Baf60c are key inducers of myocardial differentiation, Tbx5 is essential for the ability of cardiac cells to contract. [Int J Biochem Cell Biol] Abstract

    Human Adipose Tissue-Derived Mesenchymal Stem Cells Target Brain Tumor-Initiating Cells
    Both in vitro and in vivo experiments showed that human adipose tissue derived mesenchymal stem cells possess migratory abilities to target brain tumor-initiating cells isolated from medulloblastoma, atypical teratoid/rhabdoid tumors and glioblastoma. [PLoS One] Full Article

    Connexin43 Mediated Delivery of ADAMTS5 Targeting siRNAs from Mesenchymal Stem Cells to Synovial Fibroblasts
    Researchers transduced human mesenchymal stem cells (hMSCs) with ADAMTS5-targeting shRNA or non-targeted shRNA, and co-cultured them with synovial fibroblasts to allow delivery of siRNAs from hMSC to synovial fibroblasts. They found that co-culture of hMSCs-shRNA-ADAMTS5 and synovial fibroblasts reduced ADAMTS5 expression relative to co-culture of hMSCs-shRNA-control and synovial fibroblasts. [PLoS One] Full Article

    Effect of Bioactive Glass Particles on Osteogenic Differentiation of Adipose-Derived Mesenchymal Stem Cells Seeded on Lactide and Caprolactone Based Scaffolds
    In this study the influence of incorporating bioactive glass particles to lactide and caprolactone based porous scaffolds on osteogenic differentiation of adipose-derived stem cells was analyzed. [J Biomed Mater Res A] Abstract

    Multiparameter Analysis of Human Bone Marrow Stromal Cells Identifies Distinct Immunomodulatory and Differentiation-Competent Subtypes
    Scientists used immortalized human bone marrow stromal cells (BMSC) clonal lines for multi-level analysis of functional markers for BMSC subsets. All clones expressed typical BMSC cell-surface antigens; however, clones with trilineage differentiation capacity exhibited enhanced vascular interaction gene sets, whereas non-differentiating clones were uniquely CD317 positive with significantly enriched immunomodulatory transcriptional networks and high IL-7 production.     Mesenchymal Stem Cells Therapy for Autoimmune Disease: Risks and Rewards
    The authors analyze the current data from early phase trials into Crohn’s disease, systemic lupus erythematosus, and rheumatoid arthritis.     Mesenchymal Stem Cells and Adaptive Immune Responses
    The authors review the influence of mesenchymal stem cells (MSCs) on adaptive immune system, especially their bidirectional interaction in tuning the immune microenvironment and subsequently repairing damaged tissue.
