Optimization of Human Mesenchymal Stem Cell Manufacturing: The Effects of Animal/Xeno-Free Media
Human BM mesenchymal stem cells (MSC) and AdMSC were expanded in media supplemented with either human platelet lysates, serum-free media/xeno-free FDA-approved culture medium, or fetal bovine serum and the effects on their properties were investigated. [Sci Rep] Full Article

Mechanosensitive TRPM7 Mediates Shear Stress and Modulates Osteogenic Differentiation of Mesenchymal Stromal Cells through Osterix Pathway
Scientists demonstrated that intermittent fluid shear stress, a potent and physiologically relevant mechanical stimulus, regulates osteogenic differentiation of mesenchymal stromal cells through transient receptor potential melastatin 7 (TRPM7)-osterix axis. [Sci Rep] Full Article

Differentiation Capacity and Maintenance of Differentiated Phenotypes of Human Mesenchymal Stromal Cells Cultured on Two Distinct Types of 3D Polymeric Scaffolds
The differentiation potential of human mesenchymal stromal cells into the chondrogenic and osteogenic lineages on two distinct three-dimensional (3D) scaffolds, additive manufactured electrospun scaffolds, was assessed and compared to conventional two-dimensional culture. [Integr Biol] Abstract

Characterization of Secretomes Provides Evidence for Adipose-Derived Mesenchymal Stromal Cells Subtypes
Investigators deciphered the secretome of adipose-derived mesenchymal stromal cells cultured in standard and hypoxic conditions to reveal proteins, which may be responsible for regenerative action of these cells. [Stem Cell Res Ther] Abstract

Porcine Lung Mesenchymal Stromal Cells Possess Differentiation and Immunoregulatory Properties
Scientists isolated mesenchymal stem cells from the lungs (L-MSCs) of four to six-week-old germ-free pigs. They determined the self-renewal, proliferation and differentiation potential of L-MSCs. They also examined the mechanisms of immunoregulation by porcine L-MSCs. [Stem Cell Res Ther] Full Article

O-GlcNAcylation Negatively Regulates Cardiomyogenic Fate in Adult Mouse Cardiac Mesenchymal Stromal Cells
Scientists assessed the role of O-GlcNAc in differentiation of cardiac mesenchymal stromal cells. Exposure of these cells to routine differentiation protocols in culture increased markers of the cardiomyogenic lineage such as Nkx2.5 and connexin 40, and augmented the abundance of transcripts associated with endothelial and fibroblast cell fates. [PLoS One] Full Article

Bovine (Bos taurus) Bone Marrow Mesenchymal Cell Differentiation to Adipogenic and Myogenic Lineages
Scientists evaluated the effect of peroxisome proliferator-activated receptor agonists on the differentiation and metabolic features of bovine bone marrow-derived mesenchymal cells induced to adipogenic or myogenic lineages. [Cells Tissues Organs] Abstract

Repair of Segmental Mandibular Bone Defects in Sheep Using Bone Marrow Stromal Cells and Autologous Serum Scaffold. A Pilot Study
The authors evaluated effective bone regeneration using an autologous serum scaffold (alone or seeded with autologous bone marrow-mesenchymal stem cells), when implanted in a 30 mm length segmental mandibular defect in sheep. [J Clin Periodontol] Abstract

Adipose Tissue-Derived Stem Cells Reduce Acute and Chronic Kidney Damage in Mice
Investigators evaluated the effect of adipose tissue-derived stem cells in an experimental model of nephrotoxicity induced by folic acid in FVB mice. [PLoS One] Full Article

MSCs: The Sentinel and Safe-Guards of Injury
Mesenchymal stem cells (MSCs) act as sentinels to safeguard the individual from intrusion and chronic injury. A societal treatment system has evolved, paramedics and emergency room procedures, which mirror in a macro- sense what MSCs orchestrate in a micro- sense. [J Cell Physiol] Abstract

Molecular Signatures of Mesenchymal Stem Cell-Derived Extracellular Vesicle-Mediated Tissue Repair
The authors review the latest reports regarding the therapeutic potential of mesenchymal stem cell (MSC)-extracellular vesicles (EVs) by focusing on the underlying molecular mechanisms of their effects. Specifically, they feature the effects of MSC-EVs in terms of their content molecules and of the tissue recovery processes endowed by these molecules. [Stem Cell Res Ther] Full Article
