    TOP STORY
Functional Inhibition of Mesenchymal Stromal Cells in Acute Myeloid Leukemia
Addressing the contribution of mesenchymal stromal cells (MSC) to acute myeloid leukemia (AML)-induced hematopoietic failure, investigators showed that MSC from AML patients exhibit significant growth deficiency and impaired osteogenic differentiation capacity. [Leukemia] Abstract

Biophysical Regulation of Chromatin Architecture Instills a Mechanical Memory in Mesenchymal Stem Cells
Scientists identified the operative molecular mechanisms through which dynamic tensile loading regulates changes in chromatin organization and nuclear mechanics in mesenchymal stem cells. [Sci Rep] Full Article

Mesenchymal Stem Cells Can Delay Radiation-Induced Crypt Death: Impact on Intestinal CD44+ Fragments
Scientists investigated the specific roles of mesenchymal stem cells in protecting organoids against radiation-induced crypt death. When co-cultured with mesenchymal stem cells, the crypt domains of irradiated organoids possess more proliferative cells and fewer apoptotic cells than those not co-cultured with mesenchymal stem cells. Cryopreservation of Equine Mesenchymal Stem Cells in 95% Autologous Serum and 5% DMSO Does Not Alter Post-Thaw Growth or Morphology In Vitro Compared to Fetal Bovine Serum or Allogeneic Serum at 20 or 95% and DMSO at 10 or 5%
Equine bone marrow derived mesenchymal stem cells were cryopreserved in six different solutions consisting of 20% serum, 10% DMSO and 70% media or 95% serum and 5% DMSO. There were no significant differences in post-thaw viability, total cell number, morphology scores or growth kinetics among the six solutions. [Stem Cell Res Ther] Full Article

ROS Activates JNK-Mediated Autophagy to Counteract Apoptosis in Mouse Mesenchymal Stem Cells In Vitro
Investigators examined whether and how autophagy was involved in reactive oxygen species (ROS)-induced apoptosis of mesenchymal stem cells. [Acta Pharmacol Sin] Abstract

Peroxisome Proliferator-Activated Receptor Gamma Negatively Regulates the Differentiation of Bone Marrow-Derived Mesenchymal Stem Cells toward Myofibroblasts in Liver Fibrogenesis
Peroxisome proliferator-activated receptor γ (PPARγ) expression was decreased in mouse fibrotic liver. In addition, PPARγ declined during the differentiation of BMSCs toward myofibroblasts induced by transforming growth factor β1. [Cell Physiol Biochem] Full Article

Melatonin Facilitates Adipose-Derived Mesenchymal Stem Cells to Repair the Murine Infarcted Heart via the SIRT1 Signaling Pathway
The authors demonstrated that melatonin promoted functional survival of adipose derived mesenchymal stem cells (AD-MSCs) in infarcted heart and provoked a synergetic effect with AD-MSCs to restore heart function. This in vivo effect of melatonin was associated with alleviated inflammation, apoptosis, and oxidative stress in infarcted heart. [J Pineal Res] Abstract

Bone Marrow Mesenchymal Stem Cell Aggregate: An Optimal Cell Therapy for Full-Layer Cutaneous Wound Vascularization and Regeneration
Cell-aggregates constructed with green fluorescent protein-expressing rat bone marrow mesenchymal stem cells were applied to rat acute full-layer cutaneous wound model to confirm its pro-regeneration ability and compare its regenerative efficacy with the currently thriving subcutaneous and intravenous stem cell administration strategy. [Sci Rep] Full Article

Antioxidant and Anti-Inflammatory Effects of Intravenously Injected Adipose Derived Mesenchymal Stem Cells in Dogs with Acute Spinal Cord Injury
Researchers aimed to identify ways to reduce the usage and risk of high doses of glucocorticoid steroids, and determine whether AD-MSCs could be used as an early alternative treatment modality for acute spinal cord injury. [Stem Cell Res Ther] Full Article

Bioprocessing Strategies for the Large-Scale Production of Human Mesenchymal Stem Cells: A Review
Optimizing a bioprocess to generate human mesenchymal stem cells (hMSCs) or their secreted products (or both) promises to improve the efficacy as well as safety of this stem cell therapy. In this review, current media and methods for hMSC culture are outlined and bioprocess development strategies discussed. [Stem Cell Res Ther] Full Article
