    Investigators report that the mediator subunit MED23 cooperates with RUNX2 to regulate osteoblast differentiation and bone development. Med23 deletion in mesenchymal stem cells or osteoblast precursors resulted in multiple bone defects similar to those observed in Runx2+/− mice. [Nat Commun] Full Article

    Scientists investigated the molecular mechanisms underlying ATP-induced Ca2+ signaling and their roles in the regulation of cell migration in human dental pulp mesenchymal stem cells. [Stem Cells] Abstract

    Tissue-Specific Differentiation Potency of Mesenchymal Stromal Cells from Perinatal Tissues
    The authors successfully isolated and cultured mesenchymal stromal cells from amnion (AMSCs), chorion, and umbilical cord. Proliferation potential was different among them, that AMSCs revealed the lowest proliferation rate due to increased Annexin V and senescence-associated β-galactosidase positive cells.     A20 Plays a Critical Role in the Immunoregulatory Function of Mesenchymal Stem Cells
    Investigators provided new information to partly explain the molecular mechanisms of immunoregulation by mesenchymal stem cells (MSCs). Specifically, they found that A20 expression was induced in MSCs by inflammatory cytokines. [J Cell Mol Med] Full Article

    Prospective Purification of Perivascular Presumptive Mesenchymal Stem Cells from Human Adipose Tissue: Process Optimization and Cell Population Metrics across a Large Cohort of Diverse Demographics
    Researchers described an optimized protocol that aims to deliver pure, viable and consistent yields of perivascular stem cells (PSC) from adipose tissue. They analyzed the frequency of PSC within adipose tissue, and the effect of patient and procedure based variables on this yield. [Stem Cell Res Ther] Full Article

    Effect of miR-26a-5p on the Wnt/Ca2+ Pathway and Osteogenic Differentiation of Mouse Adipose-Derived Mesenchymal Stem Cells
    Investigators examined the role of miR-26a-5p in the regulation of osteogenic differentiation of mouse adipose-derived mesenchymal stem cells by using mimics and inhibitors of this microRNA. They showed that over-expression of miR-26a-5p inhibited osteogenesis and that suppression of endogenous miR-26a-5p promoted osteogenesis. [Calcif Tissue Int] Abstract

    α2-6Sialylation Is a Marker of the Differentiation Potential of Human Mesenchymal Stem Cells
    Researchers used lectin microarray technology to identify cell surface glycans as markers of the differentiation potential of stem cells. They found that α2-6Sia-specific lectins showed stronger binding to early passage adipose-derived human mesenchymal stem cells than late passage cells. [Glycobiology] Abstract

    Hypoxia Enhances Tenocyte Differentiation of Adipose-Derived Mesenchymal Stem Cells by Inducing Hypoxia-Inducible Factor-1α in a Co-Culture System
    Scientists investigated effects of O2 tension and potential signaling pathways on adipose-derived mesenchymal stem cells differentiation into tenocytes, in an indirect co-culture system. [Cell Prolif] Abstract

    Mitochondrial Transfer via Tunneling Nanotubes (TNT) is an Important Mechanism by which Mesenchymal Stem Cells Enhance Macrophage Phagocytosis in the In Vitro and In Vivo Models of ARDS
    The authors investigated the hypothesis that the anti-microbial effect of mesenchymal stromal cells in vivo depends on their modulation of macrophage phagocytic activity which occurs through mitochondrial transfer. [Stem Cells] Abstract

    Icariin Attenuates Titanium-Particle Inhibition of Bone Formation by Activating the Wnt/β-Catenin Signaling Pathway In Vivo and In Vitro
    Researchers demonstrated that icariin significantly attenuated titanium-particle inhibition of osteogenic differentiation of mesenchymal stem cells. Additionally, icariin increased bone mass and decreased bone loss in titanium-particle-induced osteolytic sites. [Sci Rep] Full Article

    Human Embryonic Stem Cell-Derived Mesenchymal Stromal Cells Ameliorate Collagen-Induced Arthritis by Inducing Host-Derived Indoleamine 2,3 Dioxygenase
    Scientists tested the therapeutic potential of human pluripotent stem cell-mesenchymal stromal cells (hPSC-MSC) of embryonic origin in the experimental model of collagen-induced arthritis. Mice were treated with either a single dose of hESC-MSC on the day of immunization or with three doses of hESC-MSC every other day starting on the day of arthritis onset.
