TOP STORYThe authors report that the microtubule plus-end tracking protein SLAIN2, which suppresses catastrophes, was not required for 2D cell migration but is essential for mesenchymal cell invasion in 3D culture and in a mouse cancer model. [Dev Cell] Abstract | Graphical Abstract

PUBLICATIONS(Ranked by impact factor of the journal)IN VITRORegionally Restricted Hox Function in Adult Bone Marrow Multipotent Mesenchymal Stem/Stromal Cells Investigators showed that Hox11 paralogous genes, which developmentally pattern the zeugopod (radius/ulna and tibia/fibula), remain regionally expressed in the adult skeleton. Using Hoxa11EGFP reporter mice, they demonstrated expression exclusively in multipotent mesenchymal stromal cells in the bone marrow of the adult zeugopod. Hox-positive cells express PDGFRα and CD51, are marked by LepR-Cre, and exhibit colony-forming unit fibroblast activity and tri-lineage differentiation in vitro. [Dev Cell] Abstract | Graphical Abstract Scientists showed that human adipose mesenchymal stem cells-derived conditioned medium exhibited inhibitory effects on A2780 human ovarian cancer cells by blocking the cell cycle, and activating mitochondria-mediated apoptosis signaling. [Sci Rep] Full Article Scientists genetically modified equine adipose-derived stem cells with endothelial nitric oxide synthase (eNOS), CAV-1WT, and a CAV-1F92A (CAV-1WT mutant) and assessed nitric oxide-mediated osteogenic differentiation and the relationship with the Wnt signaling pathway. [Stem Cell Res Ther] Full Article Researchers investigated the combined effect of substratum nano-topography and mechanical stiffness in directing mesenchymal stem cell chondrogenesis. [Tissue Eng Part A] Abstract Scientists showed that mesenchymal stromal cells (MSCs) reside in the tumor microenvironment of patients with oral cavity and oral pharyngeal squamous cell carcinoma (SCC) and are recruited via paracrine mediated tumor cell secretion of platelet derived growth factor PDGF-AA. The MSC markers CD90+, CD105+, and gremlin-1+ were found to co-localize on cells within the tumor microenvironment in oral cavity SCC specimens distinct from α-smooth muscle actin staining cancer associated fibroblasts. [J Transl Med] Full Article IN VIVOInvestigators aimed to enhance the immunomodulatory properties of mesenchymal stem cells (MSCs) via continuous secretion of the anti-inflammatory cytokine interleukin-13 (IL-13). By using MSCs as carriers of IL-13 (MSC/IL-13), they investigated their therapeutic potential, compared with non-engineered MSCs, in a mouse model of spinal cord injury (SCI). They showed that transplanted MSC/IL-13 significantly improve functional recovery following SCI, and also decrease lesion size and demyelinated area by more than 40%. [Stem Cell Reports] Full Article | Graphical Abstract Scientists evaluated the osteogenic effects of dense collagen gel scaffolds seeded with mesenchymal dental pulp stem cells on bone regeneration in a rat critical-size calvarial defect model. [Sci Rep] Full Article Researchers investigated the effects of cartilaginous extracellular matrix components and culture regimen on cartilage formation by self-assembling human mesenchymal stem cells in vitro and in vivo. [Stem Cell Res Ther] Full Article Researchers evaluated the effects of poor maternal nutirtion during gestation on bone and mesenchymal stem cell function in offspring. [PLoS One] Full Article

INDUSTRY NEWSEMBO Installation Grants to Support Ten Researchers in Establishing Laboratories Ten life scientists have been awarded EMBO Installation Grants to set up independent research laboratories in the Czech Republic, Estonia, Poland, Portugal and Turkey. Installation Grants are awarded annually. They are funded primarily by the participating member states Czech Republic, Estonia, Poland, Portugal and Turkey. Grantees are selected by a committee of EMBO Members on the basis scientific excellence as the primary selection criterion. [EMBO] Press Release Regenexx® Releases Patient Outcomes Application for Orthopedic Stem Cell Treatments Regenexx launched a new patient outcomes application. Their new app allows easily viewable results of what all participating Regenexx patients, not just a selected few, report regarding their outcomes following treatment. [Regenexx [PR Newswire Association LLC.]] Press Release Bayer AG and Versant Ventures announced the launch of BlueRock Therapeutics, a next-generation regenerative medicine company that plans to develop best-in-class induced pluripotent stem cell therapies to cure a range of diseases using an industry-leading platform. One of BlueRock Therapeutics’ initial programs is to regenerate heart muscle in patients who have had a heart attack or are suffering from chronic heart failure. [Bayer Inc.] Press Release

POLICY NEWSUnder 21st Century Cures Legislation, Stem Cell Advocates Expect Regulatory Shortcuts As the 21st Century Cures Act cleared the U.S. Senate, many attendees of the annual World Stem Cell Summit took a victory lap. The meeting assembled some of the lawyers, analysts, and activists who have long pushed for reform of how the U.S. Food and Drug Administration handles regenerative medicine. And the behemoth biomedical bill predicted to get a presidential signature could allow some stem cell products a faster and more flexible premarket approval process. [ScienceInsider] Editorial Brexit by the Numbers: The Fear of Brain Drain British universities fear losing large swathes of their research staff as the country faces up to Brexit, the split with the European Union. More than 31,000 academics at UK universities are non-British EU citizens, and so may lose their rights to live in the United Kingdom after Brexit. [Nature Trend Watch] Editorial Italian Scientists Won’t Miss Departing Prime Minister Matteo Renzi Italian politics is in turmoil after the resignation of Prime Minister Matteo Renzi — but researchers say that they are not particularly sad to see him go. In his almost three years in charge, Renzi promised improvements for universities and science but failed to raise the status of research in the country, according to scientists who complain that he also directly interfered in academic affairs.
