CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) DIABETES Generation of Rat Pancreas in Mouse by Interspecific Blastocyst Injection of Pluripotent Stem Cells Mouse wild-type pluripotent stem cells (PSCs) injected into Pdx1-/- (pancreatogenesis-disabled) mouse blastocysts developmentally compensated vacancy of the pancreatic “developmental niche,” generating an almost entirely PSC-derived pancreas. [Cell] Novel Small Molecule Glucagon-Like Peptide-1 Receptor Agonist Stimulates Insulin Secretion in Rodents and from Human Islets The studies characterize the insulinotropic effects of an early-stage, small molecule glucagon-like peptide-1 receptor agonist and provide compelling evidence to support pharmaceutical optimization. [Diabetes] Insulin Reciprocally Regulates Glucagon Secretion in Humans Researchers document that insulin is a beta cell secretory product that, in concert with glucose and among other signals, reciprocally regulates alpha cell glucagon secretion in humans. [Diabetes] Human-IAPP Disrupts the Autophagy/Lysosomal Pathway in Pancreatic Beta Cells: Protective Role of p62-Positive Cytoplasmic Inclusions Researchers report that increased expression of human-islet amyloid polypeptide (IAPP) led to impaired autophagy, due at least in part to the disruption of lysosome-dependant degradation. [Cell Death Differ] Declining Beta Cell Function Relative to Insulin Sensitivity With Increasing Fasting Glucose Levels in the Nondiabetic Range in Children The impairment in beta cell function relative to insulin sensitivity is apparent even within the nondiabetic fasting plasma glucose range in children. [Diabetes Care] Common Genetic Variation in GLP1R and Insulin Secretion in Response to Exogenous GLP-1 in Nondiabetic Subjects Variation in GLP1R may alter insulin secretion in response to exogenous glucagon-like peptide (GLP)-1. [Diabetes Care] Neonatal Diabetes Caused by Mutations in Sulfonylurea Receptor 1: Interplay between Expression and Mg-Nucleotide Gating Defects of ATP-Sensitive Potassium Channels Results demonstrate that E208K and V324M, located in distinct domains of SUR1, enhance transduction of Mg-nucleotide stimulation from the SUR1 nucleotide binding folds to Kir6.2. [J Clin Endocrinol Metab] PANCREATIC CANCER PTEN Loss Accelerates KrasG12D-Induced Pancreatic Cancer Development Data support a dosage-dependent role for PTEN, and the resulting dysregulation of the PI3K/AKT signaling axis, in both pancreatic ductal adenocarcinoma initiation and progression, and shed additional light on the signaling mechanisms that lead to the development of acinar-to-ductal metaplasia and subsequent malignant pancreatic intraepithelial neoplasia and pancreatic cancer. [Cancer Res] Gli3 Mediates Cell Survival and Sensitivity to Cyclopamine in Pancreatic Cancer Knockdown of GLI3 using siRNAs increased sensitivity to cyclopamine. In addition, GLI3 siRNAs decreased pancreatic adenocarcinoma cell viability and reduced expression of genes involved in hedgehog signaling (Patched 1 and GLI1) and cell proliferation, similar to cyclopamine. [Cancer Biol Ther] The Role of Autophagy in the Treatment of Pancreatic Cancer with Gemcitabine and Ionizing Radiation Results suggest that autophagy can be activated by gemcitabine and/or ionizing radiation in the treatment of pancreatic cancer cells and that activated autophagy plays a role in cancer suppression.
