Overexpression of Gjb4 Impairs Cell Proliferation and Insulin Secretion in Primary Islet Cells

Overexpression of gap junction beta 4 protein (Gjb4) in primary islet cells significantly inhibited the proliferation by 47%, reduced insulin secretion of primary islets (46%) and INS-1 cells (51%) and enhanced the rate of apoptosis by 63% in INS-1 cells.

Multiplexing DNA Methylation Markers to Detect Circulating Cell-Free DNA Derived from Human Pancreatic Beta-Cells

Scientists describe an ultrasensitive assay for detection of a β-cell-specific DNA methylation signature, by simultaneous assessment of six DNA methylation markers, that identified β-cell DNA in mixtures containing as little as 0.03% β-cell DNA (less than one β-cell genome equivalent).

GABA Stimulates β-Cell Proliferation through the mTORC1/p70S6K Pathway, an Effect Amplified by Ly49, a Novel GABAA-R Positive Allosteric Modulator

Researchers demonstrated that GABA stimulated β‐cell proliferation via the PI3K/mTORC1/p70S6K pathway in both mouse and human islets.

The Melatonin Receptor Antagonist Luzindole Induces the Activation of Cellular Stress Responses and Decreases Viability of Rat Pancreatic Stellate Cells

Investigators examined the effects of luzindole, a melatonin receptor‐antagonist, on cultured pancreatic stellate cells. Stimulation of cells with luzindole evoked a slow and progressive increase in intracellular free Ca2+ towards a plateau.

    Abstract

Epigenetic Silencing of AATK in Acinar to Ductal Metaplasia in Murine Model of Pancreatic Cancer

By comparing early mouse pancreatic lesions, non-invasive regions showed apoptosis-associated tyrosine kinase (AATK) co-expression in cells with acinar-to-ductal metaplasia, nuclear VAV1 localization, and cell cycle suppression.

VentX Expression in Tumor Associated Macrophages Promotes Phagocytosis and Immunity against Pancreatic Cancers

Investigators found that the expression of homeobox protein VentX, a master regulator of macrophage plasticity, was significantly decreased in the pancreatic ductal adenocarcinoma (PDA)-tumor associated macrophages (TAMs). They demonstrated that VentX was required for phagocytosis and that restoration of VentX expression in PDA-TAMs promoted phagocytosis through regulating the signaling cascades involved in the process.

Identification of a Novel IL-5 Signaling Pathway in Chronic Pancreatitis and Crosstalk with Pancreatic Tumor Cells

The expression of IL-5R and the effects of IL-5 were analyzed in human and murine tumor cells. IL-5 stimulated tumor cell migration and activation through STAT5 signaling, thereby suggesting an unreported tumor-promoting role for IL-5Rα in pancreatic cancer.

A Synergic Approach to Enhance Long-Term Culture and Manipulation of MiaPaCa-2 Pancreatic Cancer Spheroids

In the field of pancreatic cancer, the MiaPaCa-2 cell line is an interesting model for research but it is known for its difficulty to form stable spheroids; also, when formed, spheroids from this cell line are weak and arduous to manage and to harvest for further analyses such as multiple staining and imaging. The authors compared different methods to evaluate which one allowed to better overpass these limitations.

Genome-Wide DNA Methylation Analysis of KRAS Mutant Cell Lines

Scientists analyzed the basal CpG methylation of 11 KRAS-mutant and dependent pancreatic cancer cell lines and observed strikingly similar methylation patterns. KRAS knockdown resulted in unique methylation changes with limited overlap between each cell line.

Rapid Cancer Diagnosis and Early Prognosis of Metastatic Risk Based on Mechanical Invasiveness of Sampled Cells

Investigators evaluated the mechanical invasiveness of established breast and pancreatic cell lines and of tumor-cells from fresh, suspected pancreatic cancer tumors. The mechanical invasiveness matched the in vitro metastatic potential in cell lines as determined with Boyden chamber assays.

Insulin Receptor Endocytosis in the Pathophysiology of Insulin Resistance

The authors review the molecular mechanism of insulin receptor endocytosis and its impact on the pathophysiology of insulin resistance, and discuss the potential of Src homology phosphatase 2 as a therapeutic target for type 2 diabetes.
