A Cell-Autonomous Signature of Dysregulated Protein Phosphorylation Underlies Muscle Insulin Resistance in Type 2 Diabetes

Investigators developed an in vitro disease-in-a-dish model using iPSCs from type 2 diabetes (T2D) patients differentiated into myoblasts (iMyos). They found that T2D iMyos in culture exhibited multiple defects mirroring human disease, including an altered insulin signaling, decreased insulin-stimulated glucose uptake, and reduced mitochondrial oxidation.

Islet Transplantation in the Subcutaneous Space Achieves Long-Term Euglycemia in Preclinical Models of Type 1 Diabetes

The authors report the successful subcutaneous transplantation of pancreatic islets admixed with a device-free islet viability matrix, resulting in long-term euglycemia in diverse immune-competent and immuno-incompetent animal models.

Hypothalamic Perineuronal Net Assembly Is Required for Sustained Diabetes Remission Induced by Fibroblast Growth Factor 1 in Rats

Scientists identified extracellular matrix structures as previously unrecognized participants in the mechanism underlying diabetes remission induced by the central action of fibroblast growth factor 1.

Immunotherapy via PD-L1–Presenting Biomaterials Leads to Long-Term Islet Graft Survival

Investigators implemented a synthetic biomaterial platform for the local delivery of a chimeric streptavidin/programmed cell death-1 (SA-PD-L1) protein to direct “reprogramming” of local immune responses to transplanted pancreatic islets.

Generation and Characterization of CRISPR/Cas9-Mediated MEN1 Knockout BON1 Cells: A Human Pancreatic Neuroendocrine Cell Line

Researchers used the polyclonal human pancreatic neuroendocrine tumor cell line BON1, which expresses menin, serotonin, chromogranin A and neurotensin, to generate a monoclonal stable MEN1 knockout BON1 cell line by CRISPR/Cas9 editing.

L-Arginine Prevents Cereblon-Mediated Ubiquitination of Glucokinase and Stimulates Glucose-6-Phosphate Production in Pancreatic β-Cells

Scientists sought to determine a mechanism by which L-arginine increased glucose-stimulated insulin secretion in β-cells by finding a protein with affinity to L-arginine using arginine-immobilized magnetic nanobeads technology.

MYEOV Increases HES1 Expression and Promotes Pancreatic Cancer Progression by Enhancing SOX9 Transactivity

Researchers provided evidence of essential roles of myeloma overexpressed (MYEOV) in the development and progression of pancreatic ductal adenocarcinoma. In tumor specimens derived from pancreatic cancer patients, MYEOV was overexpressed and associated with poor prognosis.

RET Isoforms Contribute Differentially to Invasive Processes in Pancreatic Ductal Adenocarcinoma

The authors generated total RET and isoform-specific knockdown pancreatic ductal adenocarcinoma (PDAC) cell lines and assessed the contributions of RET isoforms to PDAC invasive spread. They showed that RET activity induced cell polarization and actin remodeling through activation of CDC42 and RHOA GTPases to promote directional motility in PDAC cells.

    Abstract

Aberrant Expression of a Novel Circular RNA in Pancreatic Cancer

Investigators performed a large-scale circRNA profiling analysis in human pancreatic ductal adenocarcinoma (PDAC) tissues, using circular RNA-specific RNA sequencing. They identified more than 40,000 previously unknown circRNAs, some of which were upregulated in PDAC tissues, compared with normal pancreatic tissues.
