    TOP STORY

Adipocyte Reprogramming by the Transcriptional Coregulator GPS2 Impacts Beta Cell Insulin Secretion

Researchers demonstrated that expression of the transcriptional coregulator GPS2 in white adipose tissue was correlated with insulin secretion rate in humans.

A 12‐Lipoxygenase‐Gpr31 Signaling Axis is Required for Pancreatic Organogenesis in the Zebrafish

The authors leveraged the strengths of the zebrafish—genetic manipulation and pharmacologic inhibition—to interrogate the role of 12‐lipoxygenase (12‐LOX) in pancreatic development. Lipidomics analysis during zebrafish development demonstrated that 12‐LOX‐generated metabolites of arachidonic acid increased sharply during organogenesis stages, and that this increase was blocked by morpholino‐directed depletion of 12‐LOX.

Pancreatic β-Cells Respond to Fuel Pressure with an Early Metabolic Switch

Scientists present a study of β-cell metabolism under increased fuel pressure using a stable isotope resolved NMR approach to investigate early metabolic events leading up to β-cell dysfunction.

Integrin and Autocrine IGF2-Pathways Control Fasting Insulin Secretion in β-Cells

Investigators observed a cooperation between different AKT isoforms and FAK-dependent adhesion signaling, which controlled either glucose-stimulated insulin secretion or prevented insulin secretion under fasting conditions.

Hypoxia Inducible Factor-1 Mediates Pancreatic β-Cell Dysfunction by Intermittent Hypoxia

Researchers demonstrated that hypoxia-inducible factor-1-mediated transcriptional activation of NOX4 and the ensuing increase in H2O2 contributed to intermittent hypoxia-induced pancreatic β-cell dysfunction.

    Abstract

Citrus Flavanone Metabolites Protect Pancreatic-β Cells Under Oxidative Stress Induced by Cholesterol

Cholesterol reduced pancreatic β-cell viability in a dose and time-dependent manner, with an improvement in the presence of the metabolites. Flavanone metabolites attenuated oxidative stress by reducing levels of lipid peroxides, superoxide anions, and hydrogen peroxide.

    Abstract

Myricetin Protects Pancreatic β-Cells from Human Islet Amyloid Polypeptide (hIAPP) Induced Cytotoxicity and Restores Islet Function

Myricetin supplementation inhibited human islet amyloid polypeptide aggregation and disaggregated preformed fibrils into non-toxic species. This protection was accompanied by inhibition of oxidative stress, reduction in lipid peroxidation and the associated membrane damage and restoration of mitochondrial membrane potential in INS-1E cells.

    Abstract

Human T Cells Expressing a CD19 CAR-T Receptor Provide Insights into Mechanisms of Human CD19-Positive β Cell Destruction

Coculturing CD19-expressing β-like cells and CD19 CAR-T cells resulted in T cell-mediated β-like cell death with release of activated T cell cytokines.

Systemic Muscle Wasting and Coordinated Tumor Response Drive Tumorigenesis

Scientists used a Drosophila model of high-sugar diet-enhanced tumorigenesis to uncover a systemic host-tumor metabolic circuit that supported tumor growth.

Long Noncoding RNA TSLNC8 Enhances Pancreatic Cancer Aggressiveness by Regulating CTNNB1 Expression via Association with HuR

Functional experiments demonstrated that tumor suppressor long noncoding RNA on chromosome 8p12 (TSLNC8) promoted pancreatic cancer (PC) cells’ proliferation and invasion in vitro, and enhanced PC growth and metastasis in vivo.
