Expression of miRNA-29 in Pancreatic β Cells Promotes Inflammation and Diabetes via TRAF3

Scientists report a key β cell-macrophage crosstalk pathway mediated by the miRNA-29-TNF-receptor-associated factor 3 (TRAF3) axis.

Pancreatic Islets Accumulate Cadmium in a Rodent Model of Cadmium-Induced Hyperglycemia

The authors examined the role of pancreatic islets in Cadmium (Cd)-induced hyperglycemia. They showed that within whole pancreatic tissue, Cd selectively accumulated in pancreatic islets and caused altered islet function that likely contributed to dysglycemia.

Visualization of Stem Cell Activity in Pancreatic Cancer Expansion by Direct Lineage Tracing with Live Imaging

Using genetic lineage tracing with a dual-recombinase system and live imaging, investigators showed that Dclk1+ tumor cells continuously provided progeny cells within pancreatic intraepithelial neoplasia, primary and metastatic PDAC, and PDAC-derived spheroids in vivo and in vitro.

S100A16 Promotes Metastasis and Progression of Pancreatic Cancer through FGF19-Mediated AKT and ERK1/2 Pathways

Researchers demonstrated that S100A16 promoted PDAC cell proliferation, migration, invasion, and metastasis both in vitro and in vivo. Knockdown of S100A16 induced PDAC cell cycle arrest in the G2/M phase and apoptosis.

Novel Nonsecosteroidal Vitamin D Receptor Modulator Combined with Gemcitabine Enhances Pancreatic Cancer Therapy through Remodeling of the Tumor Microenvironment

Investigators report the design and synthesis of 57 nonsecosteroidal vitamin D receptor (VDR) modulators based on the skeleton of phenyl-pyrrolyl pentane. Compounds C4, I5, and I8 exhibited excellent VDR affinity and effective inhibition of the activation of pancreatic stellate cells (PSCs), as well as potent suppression of the interaction between cancer cells and PSCs in vitro.

The Expression Profile and Textural Characteristics of C595-Reactive MUC1 in Pancreatic Ductal Adenocarcinoma for Targeted Radionuclide Therapy

Scientists determined the feasibility of Mucin 1 (MUC1)-based targeted radionuclide therapy for PDAC, by evaluating the expression profile of MUC1 in different pancreatic cells and tissues using the C595 antibody.

Deciphering the Genomic and lncRNA Landscapes of Aerobic Glycolysis Identifies Potential Therapeutic Targets in Pancreatic Cancer

An integrated analysis of transcriptional profiles revealed many differentially expressed long non-coding RNAs (lncRNAs) involved in PDAC aerobic glycolysis.

Extracellular Matrix Composition Modulates the Responsiveness of Differentiated and Stem Pancreatic Cancer Cells to Lipophilic Derivate of Gemcitabine

Researchers analyzed 4-(N)-stearoyl-gemcitabine cytotoxic activity, intracellular uptake, apoptosis, necrosis, and autophagy induction in both Panc1 cell line and their derived cancer stem cells.

Dysfunctional EGFR and Oxidative Stress-Induced PKD1 Signaling Drive Formation of DCLK1 Pancreatic Stem Cells

While PanIN cells expressed oncogenic KRas and have increased activity of epidermal growth factor receptor (EGFR), the authors demonstrated that in DCLK1+ PanIN cells EGFR signaling was not propagated to the nucleus.

    Abstract

Pharmacological Targeting of Endoplasmic Reticulum Stress in Pancreatic Beta Cells

Unresolved chronic endoplasmic reticulum (ER) stress is a major contributor to beta-cell loss of function and identity. Restoring ER homeostasis, enhancing ER-associated degradation of misfolded protein, and boosting chaperoning activity, are emerging therapeutic approaches for diabetes treatment.

Type I Interferons in Pancreatic Cancer and Development of New Therapeutic Approaches

The authors focus on the pre-clinical and clinical evidence of novel ways to take advantage of the type I IFN pathway, such as IFN based conjugates and activation of the STING and RIG-I pathways.
