    TOP STORY
Aging-Dependent Demethylation of Regulatory Elements Correlates with Chromatin State and Improved β Cell Function
The authors performed a comprehensive epigenomic analysis of the pancreatic β cell, key player in glucose homeostasis, in adolescent and very old mice. They observed a global methylation drift resulting in an overall more leveled methylome in old β cells. [Cell Metab] Abstract | Graphical Abstract

    DIABETES & PANCREATITIS

Identification of the Interaction of VP1 with GM130 which May Implicate in the Pathogenesis of CVB3-Induced Acute Pancreatitis
Researchers identified the host factor Golgi matrix protein 130 (GM130) as a novel target of coxsackievirus B3 (CVB3) during CVB3-induced acute pancreatitis. [Sci Rep] Full Article

Downregulation of Long Noncoding RNA Meg3 Affects Insulin Synthesis and Secretion in Mouse Pancreatic Beta Cells
Investigators explored the relationship between mouse maternal expressed gene 3 (Meg3) and the function of mouse beta cells in vitro and in vivo. [J Cell Physiol] Abstract

Primary Outgrowth Cultures Are a Reliable Source of Human Pancreatic Stellate Cells
Investigators examined whether outgrowth cultures from human surgical specimens exhibited reproducible phenotypic and functional characteristics of pancreatic stellate cells. [Lab Invest] Abstract

    PANCREATIC CANCER

Myoferlin Plays a Key Role in VEGFA Secretion and Impacts Tumor-Associated Angiogenesis in Human Pancreas Cancer
Researchers showed that myoferlin silencing in BxPC-3 human pancreatic cancer cells resulted in the inhibition of cell proliferation in vitro and in a significant reduction of the tumor volume in chick chorioallantoic membrane assay. [Int J Cancer] Abstract

Prostaglandin E2 Activates the mTORC1 Pathway through an EP4/cAMP/PKA and EP1/Ca2+-Mediated Mechanism in the Human Pancreatic Carcinoma Cell Line PANC-1
In PANC-1 human pancreatic cancer cells, scientists showed that PGE2 stimulated mammalian target of rapamycin complex 1 (mTORC1) activity independently of Akt, as evaluated by downstream signaling events. [Am J Physiol Cell Physiol] Abstract

Characterization of Novel Carcinoma Cell Lines for the Analysis of Therapeutical Strategies Fighting Pancreatic Cancer
The murine pancreatic adenocarcinoma cell lines 6606PDA and 7265PDA were compared to the 6606l cell line isolated from a liver metastasis from mice suffering from pancreatic cancer. In tissue culture 6606PDA and 6606l proliferated faster than 7265PDA. [Cell Biosci] Full Article

PEBP4 Promoted the Growth and Migration of Cancer Cells in Pancreatic Ductal Adenocarcinoma
Investigators found that the messenger RNA and protein level of phosphatidylethanolamine-binding protein 4 (PEBP4) was elevated in pancreatic ductal adenocarcinoma (PDAC) samples. Forced expression of PEBP4 in PDAC cell lines promoted cell growth and migration, while downregulation of PEBP4 in PDAC cells by RNA interference (RNAi) inhibited the growth, migration, and metastasis of the cancer cells. [Tumour Biol] Abstract

Effects of Cerulein on Keratin 8 Phosphorylation and Perinuclear Reorganization in Pancreatic Cancer Cells: Involvement of Downregulation of Protein Phosphatase 2A and Alpha4
Researchers found that cerulein dose-dependently induced keratin 8 phosphorylation and perinuclear reorganization in PANC-1 cells, thus leading to migration and invasion. [Environ Toxicol] Abstract

Pax4 Acts as a Key Player in Pancreas Development and Plasticity
Comparative analysis of Pax4 and Arx mutants, as well as Arx/Pax4 double mutants, showed that islet subtype destiny is mainly directed by cross-repression of the Pax4 and Arx factors. Importantly, the ectopic expression of Pax4 in α-cells was found sufficient to induce their neogenesis and conversion into β-like cells, not only during development but also in adult rodents. [Semin Cell Dev Biol] Abstract
