TOP STORYThe ETS-Transcription Factor ETV1 Regulates Stromal Expansion and Metastasis in Pancreatic Cancer Researchers investigated the function of ETV1 in stromal expansion of pancreatic ductal adenocarcinoma (PDAC) and metastasis, as well as its effects on its downstream target Sparc, which encodes a matricellular protein found in the PDAC stroma that has been associated with invasiveness and metastasis and poor outcomes of patients. PUBLICATIONS(Ranked by impact factor of the journal)DIABETES & PANCREATITISLoss of Free Fatty Acid Receptor 2 Leads to Impaired Islet Mass and Beta Cell Survival Scientists set out to explore what role fatty acid receptor 2 (FFA2) may play in regulation of β cell mass. Interestingly, Ffar2−/− mice exhibited diminished β cell mass at birth and throughout adulthood, and increased β cell death at adolescent time points, suggesting a role for FFA2 in establishment and maintenance of β cell mass. [Sci Rep] Full Article Artesunate was found to increase the survival of SAP rats significantly when injected with 3.5% sodium taurocholate into the biliopancreatic duct in a retrograde direction, improving their pancreatic pathology and decreasing serum amylase and pancreatic lipase activities along with substantially reduced pancreatic IL-1β and IL-6 release. [Int Immunopharmacol] Abstract PANCREATIC CANCERThe authors demonstrated that withaferin-A, the biologically active withanolide extracted from Withania somnifera, significantly increased autophagosomes, but blocked the degradation of autophagic cargo by inhibiting SNARE-mediated fusion of autophagosomes and lysosomes in human pancreatic cancer cells. [Autophagy] Abstract demonstrate that MUC13 functionally interacts and activates HER2 at p1248 in pancreatic ductal adenocarcinoma (PDAC) cells, leading to stimulation of HER2 signaling cascade, including ERK1/2, FAK, AKT and PAK1 as well as regulation of the growth, cytoskeleton remodeling and motility, invasion of PDAC cells—all collectively contributing to PDAC progression. [Oncogene] Abstract Investigators developed an unbiased and in vivo target discovery approach to identify molecular vulnerabilities in low-passage and patient-derived pancreatic ductal adenocarcinoma xenografts or genetically engineered mouse model-derived allografts. [Cell Rep] Full Article | Graphical Abstract P300 Inhibition Enhances Gemcitabine-Induced Apoptosis of Pancreatic Cancer The transcriptional cofactor p300 has histone acetyltransferase activity and has been reported to participate in chromatin remodeling and DNA repair. Scientists hypothesized that targeting p300 can enhance the cytotoxicity of gemcitabine, which induces pancreatic cancer cell apoptosis by damaging DNA. [Oncotarget] Full Article Investigators report that PAUF, a soluble protein involved in pancreatic tumorigenesis and metastasis, plays a role as an enhancer of tumor-infiltrating MDSC and its functional activity. They show that PAUF enhanced the accumulation of MDSCs in the spleen and tumor tissues of PAUF-overexpressing tumor cell-injected mice. [Oncotarget] Full Article Interaction of tRNA with MEK2 in Pancreatic Cancer Cells Researchers tested the consequence of the interaction between tRNA and mitogen-activated protein kinase kinase 2 (MEK2) in pancreatic cancer cell lines. They also generated disease and drug resistance-derived MEK2 mutants to evaluate the function of the tRNA-MEK2 interaction. [Sci Rep] Full Article | |

REVIEWSThe Role of Diet and Exercise in the Transgenerational Epigenetic Landscape of T2DM The reviewers focus on the current understanding of mechanisms by which lifestyle factors affect the epigenetic landscape in type 2 diabetes mellitus and obesity. Evidence from the past few years about the potential mechanisms by which diet and exercise affect the epigenome over several generations is discussed.
