TOP STORYScientists found that rodent and human T lymphocytes released exosomes containing the miRNAs miR-142-3p, miR-142-5p, and miR-155, which could be transferred in active form to β cells favoring apoptosis. [Cell Metab] Abstract | Graphical Abstract | |

PUBLICATIONS(Ranked by impact factor of the journal)DIABETES & PANCREATITISMANF Is Required for the Postnatal Expansion and Maintenance of the Pancreatic β-Cell Mass in Mice Researchers assessed the importance of mesencephalic astrocyte-derived neurotropic factor (MANF) for the postnatal expansion of the pancreatic β-cell mass and for adult β-cell maintenance in mice. Detailed analysis of Pdx-1Cre+/-::Manffl/fl mice revealed mosaic MANF expression in postnatal pancreases and significant correlation between the number of MANF-positive β-cells and β-cell mass in individual mice. [Diabetes] Abstract Scientists investigated the molecular mechanism of pancreatic islet-derived mesenchymal stem cell differentiation into beta-cells in the presence of insulin and leptin resistance stimulators. They determined that beta-cell differentiation was stimulated by glucose, insulin, and leptin. [Biochim Biophys Acta] Abstract Investigators evaluated apoptosis, oxidative stress, autophagy and microRNA expression in multipotent intestinal epithelial stem cells and pancreatic islets isolated post mortem from healthy and EMS diagnosed horses. [Int J Mol Sci] Full Article PANCREATIC CANCERPDAC (pancreatic ductal adenocarcinoma) cells or premalignant H6c7 pancreatic ductal epithelial cells differentially expressed ΔZip2- and ATF3, relating to stronger Nrf2 activity seen in Panc1 cells and TGF-β1 activity in T3M4 or H6c7 cells, respectively. [Oncogene] Abstract Researchers showed that CD133Hi pancreatic tumor initiating cells had increased expression of mevalonic-acid-pathway enzymes, cholesterol-content and caveolin expression. [Cancer Lett] Abstract Irisin Inhibits Pancreatic Cancer Cell Growth via the AMPK-mTOR Pathway Human recombinant nonglycosylated P-irisin or glycosylated E-irisin were compared to examine the role of recombinant irisin against pancreatic cancer cell lines, MIA PaCa-2 and Panc03.27 and cell colony formation assays revealed that irisin significantly inhibited the growth of MIA PaCa-2 and Panc03.27 in a dose-dependent manner. [Sci Rep] Full Article Pancreatic cancer (PC) cells with overexpression or knockdown of vasohibin 2 (VASH2) were used to examine the involvement of VASH2 in epithelial-mesenchymal transition (EMT) by detecting the expression of epithelial and mesenchymal markers and EMT-related transcription factor ZEB1/2, in gemcitabine resistance and tumor cell invasion by apoptosis and invasion assays, and in cancer stem cell-like phenotypes by detecting the proportion of CD24+CD44+ and side population cells in PC cells with flow cytometry. [Cancer Med] Full Article miR-429 Suppresses Neurotrophin-3 to Alleviate Perineural Invasion of Pancreatic Cancer The authors investigated roles of miR-429 in modulating perineural invasion (PNI) in pancreatic ductal adenocarcinoma (PDAC). Investigators found that miR-429 was downregulated in PDAC cancer tissues and was profoundly decreased in tissues with PNI. REVIEWSThe α-Cell in Diabetes Mellitus RNA sequencing reveals that, despite their opposing roles in the control of blood levels of glucose, α-cells and β-cells have remarkably similar patterns of gene expression. This similarity might explain the fairly facile interconversion between these cells and the ability of the α-cell compartment to serve as a source of new β-cells in models of extreme β-cell loss that mimic type 1 diabetes mellitus. [Nat Rev Endocrinol] Abstract Pancreatic Islet Dysfunction in Type 2 Diabetes Mellitus Compelling evidence suggests that accumulation of islet amyloid in the islets of Langerhans significantly contribute to β-cell dysfunction and diabetes. Emerging evidence implicates a role for cystic fibrosis transmembrane-conductance regulator in the regulation of insulin secretion from pancreatic islets. INDUSTRY NEWSThe Lustgarten Foundation’s investment will support postdocs, graduate students, technicians and a senior scientist for its duration. The Lustgarten Lab’s goals are to better understand the immunological conditions and genetic events that contribute to the development of pancreatic cancer, to study the disease on a single-cell level in both humans and mouse models, and to develop novel high throughput tools for culture and drug testing using mini-organs known as organoids. [Massachusetts Institute of Technology] Press Release Rice U. Pursues End Game for Diabetes The National Institutes of Health has awarded Omid Veiseh’s lab a prestigious four-year, $2.8 million R01 grant to design hydrogel-encapsulated cells that, when placed into a patient, sense blood glucose levels and produce insulin on demand. [Rice University] Press Release Helix BioPharma Corp. announced that it has extended its collaboration agreement with Moffitt Cancer Center for an additional year. During the first year of the collaboration, a new pancreatic adenocarcinoma mouse model suitable for testing L-DOS47 in combination with immunotherapy was developed. [Helix BioPharma Corp.
