    TOP STORY
Is Metastatic Prostate Cancer Tailor-Made for Precision Oncology?
Researchers showed that though metastases from different patients varied widely in their genetic characteristics, metastases within a single patient were remarkably similar. These results suggest not merely that patients with metastatic prostate cancer may benefit from treatment tailored to their particular tumors, but also that a single biopsy may provide enough information to oncologists to guide such therapy. LABORATORY RESEARCH

Stem Cell and Neurogenic Gene-Expression Profiles Link Prostate Basal Cells to Aggressive Prostate Cancer
The authors described a genome-wide transcriptome analysis of human benign prostatic basal and luminal epithelial populations using deep RNA sequencing. Through molecular and biological characterizations, they showed that the differential gene-expression profiles account for their distinct functional properties. High Throughput Microscopy Identifies Bisphenol AP, a Bisphenol a Analog, as a Novel AR Down-Regulator
Researchers used high throughput microscopy and quantitative image analysis to evaluate effects of selected endocrine disruptors on androgen receptor (AR) levels in multiple breast and prostate cancer cell lines. Bisphenol AP (BPAP), which is used in chemical and medical industries, was identified as a down-regulator of both full length AR and the AR-V7 splice variant. [Oncotarget] Full Article

Roles of Distal and Genic Methylation in the Development of Prostate Tumorigenesis Revealed by Genome-Wide DNA Methylation Analysis
Investigators performed a MBD-seq analysis on prostate specimens classified into low, high, and very high risk group based on Gleason score and TNM stages. [Sci Rep] Full Article

Triiodthyronine Attenuates Prostate Cancer Progression Mediated by β-Andregenic Stimulation
Scientists analyzed the effects of β-adrenergic stimulation, and/or thyroid hormones on neuroendocrine differentiation and cell invasion, using in vivo (LNCaP tumor) and in vitro models (LNCaP and DU145 human cells). [Mol Med] Full Article

Beta-Arrestin-2 Counters CXCR7-Mediated EGFR Transactivation and Proliferation
The authors used cDNA knock-in, RNA interference and analysis of mitogenic signaling components in both normal prostate epithelial cells and prostate cancer cells to decipher the proliferation inducing mechanism of the atypical 7-transmebrane chemokine receptor (CXCR7)-epidermal growth factor receptor (EGFR) interaction. [Mol Cancer Res] Abstract

Stroma-Induced Jagged1 Expression Drives PC3 Prostate Cancer Cell Migration; Disparate Effects of RIP-Generated Proteolytic Fragments on Cell Behaviour and Notch Signaling
Investigators showed that bone marrow stroma induces Jagged1 expression in bone metastatic prostate cancer PC3 cells and that this enhanced expression is mechanistically linked to the promotion of cell migration. [Biochem Biophys Res Commun] Abstract

    CLINICAL RESEARCH

Common Variation in BRCA1 May Have a Role in Progression to Lethal Prostate Cancer after Radiation Treatment
Researchers found that two single-nucleotide polymorphisms, rs4473733 and rs8176305, were associated with lethal prostate cancer in men receiving radiation therapy. [Prostate Cancer Prostatic Dis] Abstract

Vimentin and Ki67 Expression in Circulating Tumor Cells Derived from Castrate-Resistant Prostate Cancer
Researchers asked if the expression of vimentin and Ki67 in circulating tumor cells offered prognostic or predictive information in castrate-resistant prostate cancer. [BMC Cancer] Full Article

New Insights into the Androgen Biotransformation in Prostate Cancer: A Regulatory Network Among Androgen, Androgen Receptors and UGTs
Although many inhibitors of the enzymes responsible for the synthesis of androgens have been developed into drugs to fight against prostate cancer, the elimination procedures metabolized by the UDP-glucuronyltransferases (UGTs) are less emphasized. Thus the regulatory network among androgen, androgen receptors and UGTs is carefully reviewed in this article, indicating the determinant effects of UGTs on prostatic androgens and the regulation of androgen receptors. INDUSTRY NEWS
Prostate Cancer Vaccine Trial Seeks Volunteers
In an initial study led by Professor Freddie Hamdy and Professor Jim Catto, researchers will vaccinate 48 men with low or intermediate risk prostate cancer to document the safety of the new vaccine and measure how well the immune system to responds to it. [Oxford University] Press Release

Queen’s University in World’s First Trial of New Treatment for Prostate Cancer
A new approach being trialed by Queen’s researchers is the first to combine two existing forms of radiotherapy – Volumetric Modulated Arc Therapy to target prostate cancer cells in the pelvis, along with Radium 223 to target the disease in the bones. [Queen’s University] Press Release
