The Kinase Activity of the Ser/Thr Kinase BUB1 Promotes TGF-β Signaling
Researchers identified the kinase BUB1 (budding uninhibited by benzimidazoles-1) as a key mediator of transforming growth factor-β (TGF-β) signaling. A small-molecule inhibitor of the kinase activity of BUB1 administration to mice bearing lung carcinoma xenografts reduced the amount of phosphorylated SMAD2 in tumor tissue. PKCδ-iPLA2-PGE2-PPARγ Signaling Cascade Mediates TNF-α Induced Claudin 1 Expression in Human Lung Carcinoma Cells
Investigators elucidated the molecular mechanisms involved in TNF-α induced Claudin 1 (CLDN1) expression in human lung carcinoma A549 cells. Chemical inhibition or siRNA downregulation of Src, PI3K, Akt, MAPKs, NFκB, caspase and PKC demonstrated that PKC, specifically PKCδ, is required for TNF-α induced CLDN1 expression. A diverse panel of human lung cancer cell lines and a SHP77 small cell lung cancer xenograft model were used to investigate the anti-tumor activity of carfilzomib (CFZ).
