Researchers showed that lysosomes in aged HSCs are hyperacidic, depleted, damaged, and aberrantly activated. Single-cell transcriptomics and functional analyses reveal that suppression of hyperactivated lysosomes using a vacuolar ATPase inhibitor restores lysosomal integrity and metabolic and epigenetic homeostasis in old HSCs.
